摘 要:目的:制备具有组织黏附性的白藜芦醇聚合物胶束,并采用HPLC法测定白藜芦醇胶束中白藜芦醇的含量。方法:采用多聚赖氨酸孔板实验和小动物荧光成像法进行胶束黏附性实验,优化胶束处方获得黏附性较好的白藜芦醇胶束。以甲醇-水溶液(42∶58)为流动相,经资生堂SPOLAR C18柱(150 mm×4.6 mm,5μm)色谱柱分离,流速为1.0 mL/min,检测波长为305 nm,柱温为35°C,进样量为10μL。结果:单独以F127为辅料制备的白藜芦醇胶束黏附性最佳。白藜芦醇在10~200μg/mL浓度范围内峰面积与浓度线性关系良好(r=0.9996),专属性、精密度、回收率、稳定性均符合方法学要求。结论:本实验成功制备具有组织黏附性的白藜芦醇胶束,并建立了测定胶束中白藜芦醇含量的方法,该方法准确、快速、简便。Objective:To prepare resveratrol polymer micelles with tissue adhesion and determine the content of resveratrol in the micelles by HPLC.Methods:The micelle adhesion experiment was carried out by polylysine orifice plate experiment and small animal fluorescence imaging method,and the micelle prescription was optimized to obtain resveratrol micelles with good ad⁃hesion.The separation was performed on a Shiseido SPOLAR C18 column(150 mm×4.6 mm,5μm)with methanol⁃water(42∶58)as the mobile phase.The flow rate was 1.0 mL/min,the detection wavelength was 305 nm,the column temperature was 35℃,and the injection volume was 10μL.Results:Resveratrol micelles prepared with F127 alone had the best adhesion.The peak area and concentration of resveratrol had a good linear relationship in the concentration range of 10~200μg/mL(r=0.9996).The specificity,precision,recovery and stability all met the methodological requirements.Conclusion:In this experiment,resve⁃ratrol micelles with tissue adhesion were successfully prepared,and a method for the determination of resveratrol content in mi⁃celles was established.The method is accurate,rapid and simple.
