1株猫源长湾泥沙门菌的分离鉴定及致病性分析

摘　　要：为探究引起1例就诊宠物猫腹泻病原,揭示其生物学特性和致病性。本研究采集腹泻病猫肛门拭子,通过细菌分离培养、PCR扩增、MLST分型和血清凝集试验进行鉴定。进一步开展生长曲线测定、生化特性鉴定、运动性试验、药敏试验和小鼠致病性试验分析其生物学特性和致病性特点。结果显示,引起该宠物猫腹泻病原为长湾泥沙门菌,命名为SJ201201,ST型为ST3918,血清型为17:a,1,5:-,生长趋势与鼠伤寒沙门菌ATCC14028无显著性差异,生化反应符合沙门菌属细菌典型反应特征,在半固体琼脂中的运动性与鼠伤寒沙门菌ATCC14028无显著性差异(P≥0.05),药敏试验结果显示长湾泥沙门菌对15种抗生素药物敏感,腹腔注射感染小鼠后,10^(9),10^(8)CFU/只感染剂量在感染后24 h内小鼠全部死亡,10^(7)CFU/只感染剂量死亡率为80%,10^(6)CFU/只感染剂量死亡率为20%,发病死亡小鼠均表现不同程度的精神沉郁、被毛粗乱、眼角分泌物增多等临床症状。结果表明,长湾泥沙门菌SJ201201具有沙门菌属一般生物学特性,并且对小鼠具有一定致病性。本研究首次从国内宠物猫样本中分离获得长湾泥沙门菌,为长湾泥沙门菌的科学防控提供参考,丰富沙门菌属细菌的流行病学资料。To determine the pathogen causing pet cat diarrhea and analyze the biological properties and pathogenicity of the bacteria,cloacal swab from the diseased cat was collected.Pathogenic bacteria were isolated and cultured.PCR amplification,MLST sequencing and serum agglutination test were conducted to identify the bacteria.Growth curve,biochemical reaction,motility assay,drug sensitivity and pathogenicity experiment were performed to study the biological properties and pathogenicity of the germ.The result showed that the pathogen causing diarrhea was Salmonella Jangwani,named SJ202101.The sequence type and serotype belonged to ST3918 and 17:a,1,5:-,respectively.The isolate could grow favorably in LB broth comparing with S.typhimurium ATCC14028.Biomedical reaction was accordance with other Salmonella serovars.The motility in semi-solid agar exhibited similarly with S.typhimurium ATCC1402(P≥0.05).Antimicrobial susceptibility test suggested that the isolate was sensitive to 15 drugs.After mice were infected by intraperitoneal injection,all mice infected with 10^(9)CFU and 10^(8)CFU S.typhimurium died within 24 h,the mortality rate was 80%in the 10^(7) CFU dose group,and 20%in the 10^(6) CFU dose group.Classical symptoms including depression,exudate in the eyes were observed.In conclusion,Salmonella Jangwani was isolated from pet cat sample for the first time in China.This work provides references for scientific prevention and control of Salmonella Jangwani and epidemiology data for Salmonella spp.
