摘  要:为了评价纳米氧化锌颗粒(ZnO NPs)对17℃猪精子保存质量的影响,本试验在17℃保存猪精子稀释液中添加20,50,100,200 mg/L ZnO NPs,对保存精子质量和功能进行分析。结果显示,与对照组比较,添加50 mg/L ZnO NPs可以显著提高保存精子的总抗氧化能力(P<0.05),降低保存精子丙二醛含量(P<0.05)。50,100,200 mg/L ZnO NPs处理组精子膜完整性显著高于对照组(P<0.05),骨架蛋白和膜蛋白磷酸化水平显著上调(P<0.05),100,200 mg/L处理组精子全蛋白磷酸化水平显著上升,且定位于精子鞭毛中段和主段。ZnO NPs添加不影响cAMP/PKA信号通路。推测ZnO NPs可能通过提高抗氧化能力来有效提高猪精子活力,消除过量丙二醛,提高猪精子蛋白磷酸化水平,从而提升膜完整性。The aim of this study was to evaluate the effects of zinc oxide nanoparticles(ZnO NPs)on the quality of boar sperm preserved at 17℃.We added 20,50,100 and 200 mg/L of ZnO NPs to a boar sperm diluent preserved at 17℃to assess sperm quality and function.The results showed that ZnO NPs significantly improved the total antioxidant capacity(T-AOC)of preserved sperm(P<0.05),and reduced the malondialdehyde(MDA)content(P<0.05).The membrane integrity of 50,100 and 200 mg/L of ZnO NPs was significantly higher than the control,and the phosphorylation levels of skeleton protein and membrane protein were significantly up-regulated(P<0.05).The phosphorylation levels of total protein of 100,200 mg/L ZnO NPs groups increased significantly,which located in middle-and principal-pieces of sperm flagella.The ZnO NPs addition did not interfere with the cAMP/PKA signaling pathway.It is hypothesized that ZnO NPs may enhance membrane integrity by improving antioxidant capacity,effectively improving porcine sperm viability,eliminating excess MDA,and increasing porcine sperm protein phosphorylation levels.
