摘  要:为探讨中药复方对公鸡精液品质的影响,24只来航公鸡随机分为3组,中药复方Ⅰ、Ⅱ组和对照组,每组8只,中药复方组分别添加中药复方Ⅰ、Ⅱ,对照组饲喂基础日粮,预试期7 d,正试期70 d。分别于试验前和试验结束称量鸡体质量,并记录每日采食量;分别于正式试验61,63,65,67,69 d采集试验鸡精液,检测精子密度、活力和畸形率,通过试剂盒检测公鸡精浆抗氧化性能;试验结束后采集翅下静脉血,并处死试验鸡摘取脾脏、肝脏及睾丸计算其脏器指数,全自动血液生化分析仪检测血清生化指标,qRT-PCR检测睾丸组织AR、Bcl-2、Bax基因mRNA表达水平。结果表明,与空白对照组相比,添加中药复方Ⅰ、Ⅱ对公鸡生产性能及血清生化指标无显著影响(P>0.05);2组中药复方均能显著提高试验公鸡的精子密度(P<0.05),其中,复方Ⅱ极显著提高精子活力(P<0.01);中药复方Ⅰ、Ⅱ均能提高精浆超氧化物歧化酶(SOD)活性和谷胱甘肽(GSH)含量,其中,复方Ⅰ可显著升高精浆GSH含量(P<0.05);中药复方Ⅰ极显著提高公鸡睾丸AR基因表达水平(P<0.01),显著升高Bcl-2/Bax的比值(P<0.05),中药复方Ⅱ可显著升高公鸡睾丸Bcl-2的表达水平(P<0.05),Bax基因表达水平均无显著性差异(P>0.05)。本研究可为应用中药复方提高公鸡精液品质提供参考。In order to investigate the effect of traditional Chinese medicine(TCM)compound on the quality of chicken rooster semen,24 Leghorns were randomly divided into 3 groups,TCM compoundⅠ,Ⅱand control group,with 8 Leghorns in each group.The TCM compound groups were added with TCM compoundⅠandⅡrespectively,and the control group was fed the basic diet.The pre-test period was 7 d and the formal test period was 70 d.The chickens were weighed before and at the end of the experiment,and the daily feed intake was recorded.Semen samples were collected on days 61,63,65,67,and 69 of the formal test period,respectively,and the sperm density,motility and malformation rate were detected.The antioxidant properties of seminal pulp were detected by a commercial kit.At the end of the experiment,the blood was collected from the wing vein and all the experimental chickens were sacrificed.The spleen,liver and testes were picked to calculate the organ index,the serum biochemical indicators were analyzed by automatic blood biochemical analyzer,and the mRNA expression level of AR,Bcl-2and Baxgene in the testicular tissue were detected by qRT-PCR.The results showed that compared with the control group,the addition of TCM compoundⅠandⅡhad no significant effect on the performance and serum biochemical indexes of rooster(P>0.05).However,both groups of TCM compound could significantly increase the sperm density(P<0.05),and the TCM compoundⅡsignificantly improved sperm motility(P<0.01).Both TCM compoundⅠandⅡcould increase the activity of superoxide dismutase(SOD)and the content of glutathione(GSH),and TCM compoundⅠcould significantly increase the content of seminal glutathione(GSH)in seminal plasma(P<0.05).TCM compoundⅠcould also significantly improve the expression of ARgene in the testis(P<0.05).and the ratio of Bcl-2/Bax(P<0.05),TCM compoundⅡcould significantly increase the expression of Bcl-2in the testis(P<0.05),but there was no significant difference in Bax gene expression among all groups(P>0.05).This study can provi
