摘  要:外泌体的相关研究已经在很多领域展开,从其作为一种信号传导分子到其在很多疾病的早期诊断起重要作用,以及作为治疗药物的转载工具等方面对其进行了深入研究。通过对近年来高非酯化脂肪酸(nonestes-terified fatty acid,NEFA)情况下外泌体miRNA加剧肝脏脂代谢障碍研究现状进行总结,指出高NEFA引起的病理变化,主要是以高NEFA诱导肝细胞发生内质网应激和脂质凋亡为特征的肝脂毒性变化;归纳总结外泌体研究进展,特别是肝细胞源外泌体miRNAs在肝脏糖脂代谢及相关代谢紊乱中的作用,讨论外泌体miRNA参与高NEFA血症引起的肝脂毒性过程,阐述肝细胞源外泌体参与到肝细胞ERS-脂质凋亡通路加剧高NEFA血症引起的肝脂毒性。在现有研究基础上,对未来在肝细胞源外泌体miRNA对肝脂毒性发生过程中的研究方向作了展望,以期对酮病奶牛肝细胞源外泌体miRNAs在肝脂毒性发生过程中的作用有更深入的了解。Exosomes have been studied in many fields,ranging from their role as a signaling molecule to their important role in the early diagnosis of many diseases and as a tool for the reproduction of therapeutic drugs.By summarizing the current status of research on exosomal miRNAs exacerbating hepatic lipid metabolism disorders under high NEFA in recent years,we point out that the pathological changes caused by high NEFA are mainly hepatic lipotoxic changes characterized by high NEFA-induced endoplasmic reticulum stress and lipid apoptosis in hepatocytes;this paper summarize the progress of exosome research,especially the role of hepatocyte-derived exosomal miRNAs in hepatic glycolipid metabolism and related.Based on the existing studies,the future directions of research on hepatocyte-derived exosomal miRNAs in the development of hepatic lipotoxicity are prospected in order to gain a better understanding of the role of hepatocyte-derived exosomal miRNAs in the development of hepatic lipotoxicity in ketosis cows.
