摘  要:目的探讨松萝提取物对胶原诱导性关节炎(CIA)大鼠高迁移率族蛋白B1(HMGB1)-晚期糖基化终产物受体(RAGE)通路及自噬相关基因的影响。方法选择5周龄SPF级Wistar雌性大鼠54只,造模成功后分为空白组、模型组、羟氯喹(HCQ)组,松萝提取物低(2 mg/kg)、中(6 mg/kg)、高(54 mg/kg)剂量组,每组9只。空白组、模型组予生理盐水灌胃,其余各组进行相应药物干预。ELISA检测血清中IL-6、HMGB1水平,RT-PCR检测Beclin1、B淋巴细胞瘤-2基因(Bcl-2)、Ⅲ型磷酸肌醇3激酶(PI3K C3)mRNA表达,Western blot检测HMGB1、RAGE、Beclin1、Bcl-2、PI3K C3蛋白表达。结果与空白组比较,松萝提取物低、中、高剂量组、模型组、HCQ组血清IL-6、HMGB1水平升高,滑膜组织HMGB1、RAGE蛋白表达和Bcl-2、PI3K C3 mRNA及蛋白表达升高,Beclin1 mRNA及蛋白表达降低,差异有统计学意义(P<0.05)。与模型组比较,HCQ组和松萝提取物中、高剂量组血清HMGB1、IL-6水平升高,滑膜组织HMGB1、RAGE蛋白表达和Bcl-2、PI3K C3 mRNA及蛋白表达降低,Beclin1 mRNA及蛋白表达升高,差异有统计学意义(P<0.05)。与HCQ组比较,松萝提取物中、高剂量组上述指标无明显变化(P>0.05)。结论松萝提取物可能通过调控自噬水平,降低炎症反应。Objective To explore the effects of usnea extract on high mobility group protein B1(HMGB1)advanced stage glycosylation end-product receptor(RAGE)pathway and autophagy related genes in collagen-induced arthritis(CIA)rats.Methods A total of 54 the grade SPE Wistar female rats of 5 weeks old were selected.After successful modeling,the rats were divided into the blank group,model group,hydroxycloroquine(HCQ)group,usnea extract low-dose group(2 mg/kg),medium dose group(6 mg/kg)and high dose group(54 mg/kg),9 cases in each group.The blank group and model group were given the normal saline by gavage,and the other groups were given the corresponding drug interventions.The serum IL-6 and HMGB1 levels were detected by ELISA method,and the mRNA expressions of Beclin1,Bcl-2 and PI3K C3 were detected by RT-PCR,and the protein expressions of HMGB1,RAGE,Beclin1,Bcl-2 and PI3K C3 were detected by the Western blot method.Results Compared with the blank group,the serum HMGB1 and IL-6 levels in the usnea low-dose,medium-dose and high-dose groups,model group and HCQ group were increased,the protein expressions of HBGB1 and RAGE and Bcl-2 and PI3K C3 mRNA and protein were in-creased,the protein expression of Beclin1 mRNA was decreased,and the differences were statistically signifi-cant(P<0.05).Compared with the model group,the HMGB1 and IL-6 levels in the HCQ group and the us-nea extract medium-dose and high-dose groups were increased,the HMGB1,RAGE protein expression and Bcl-2 and PI3K C3 mRNA and protein expression were decreased,Beclin1 mRNA and protein expressions were increased,and the differences were statistically significant(P<0.05).Compared with the HCQ group,the above indicators in the usnea extract medium-dose and high-dose groups had no significant change(P>0.05).Conclusion Usnea extract could reduce the inflammatory reaction by regulating the autophage level.
