摘　　要：旨在探究阿魏酸植物甾醇酯体内外抗猪细小病毒(PPV)作用。采用CCK-8法检测阿魏酸植物甾醇酯对PK-15细胞的安全浓度,明确最佳给药浓度,并采用RT-qPCR检测病毒增殖情况,ELISA检测细胞炎症因子分泌水平,Western blot检测线粒体自噬相关蛋白的表达水平。体内试验是给豚鼠注射PPV后灌胃阿魏酸甾醇酯,检测性腺PPV含量、血清中HI抗体含量。结果显示,阿魏酸植物甾醇酯对PK-15细胞的最高安全浓度为40μmol/mL。阿魏酸植物甾醇酯呈剂量依赖性关系显著降低PPV的病毒增殖;与空白组相比,阿魏酸植物甾醇酯显著降低PPV感染PK-15诱发细胞炎症因子IL-12、IL-17、MIP-1α、TGF-β、PGK1、IL-1β、IFN-γ、IL-6、TNF-α的表达。此外,阿魏酸植物甾醇酯显著降低PPV感染PK-15诱发线粒体自噬相关蛋白ATG3、Becline1、P62、Pink1、Parkin表达,抑制细胞线粒体自噬发生。体内试验表明阿魏酸植物甾醇酯大、中剂量组有效抑制PPV复制。说明阿魏酸植物甾醇酯体内有具有良好的抗细小病毒活性。The anti-porcine parvovirus effect of ferulic acid phytosterol ester(FAPE)was investigated in vitro and in vivo.A safe concentration of FAPE for PK-15 cells was detected using CCK-8 to determine the optimal drug concentration.In addition,RT-qPCR was used to detect viral proliferation,ELISA was used to detect secretion levels of inflammatory factors,and Western blot was used to detect expression levels of conformational related proteins.In vivo,guinea pigs were treated with intragastric administration of FAPE after PPV injection,and PPV in the gonads and HI in the serum were measured.The results showed that the maximum safe concentration of sterol FAPE for PK-15 cells was 40μmol/mL.FAPE significantly reduced PPV viral proliferation in a dose-dependent manner.Compared with the control group,the FAPE significantly reduced the expression of inflammatory factors IL-12,IL-17,MIP-1α,TGF-β,PGK1,IL-1β,IFN-γ,IL-6 and TNF-αin PPV-infected PK-15 cells.In addition,FAPE significantly reduced the expression of mitophagy-related proteins ATG3,Becline1,P62,Pink1,and Parkin induced by PPV infection with PK-15 and inhibited metagenesis in cells.In vivo experiments showed that elevated and moderate doses of FAPE were effective in inhibiting PPV replication.FAPE has a superior in vivo anti-parvovirus activity.
