摘  要:【目的】分析光照对蛋鸡不同组织和不同产蛋时期ONECUT1基因的差异表达,探讨该基因对鸡产蛋性能的影响,为深入开展光照对鸡繁殖性能分子机理的研究奠定基础。【方法】选取贵州黄鸡体重相近的10周龄母鸡216只,在半舍饲养殖条件下(舍饲+运动场)随机分为自然光照组和早晚各增光1 h组,140日龄、200日龄、480日龄屠宰各4只,采集垂体、下丘脑、视网膜、输卵管和卵巢等组织,利用qRT-PCR技术检测ONECUT1基因的差异表达。【结果】ONECUT1基因在肌肉组织中表达量最高,输卵管、卵巢次之,肝脏最低。在垂体、下丘脑和卵巢组织中,ONECUT 1基因在产蛋高峰期(200日龄)的表达量显著高于开产日期(140日龄)和产蛋后期(480日龄)。垂体组织中200日龄的增光组显著高于自然光照组,输卵管中140日龄的自然光照组显著低于增光组。【结论】ONECUT1基因的表达具有时空特性,并受光照调节,与鸡产蛋性能有关。【Objective】The effect of lighting on differential expression of ONECUT1 gene in different tissue and different laying period of Guizhou yellow chicken was analyzed,and the effect of ONECUT1 gene on laying characteristics was explored,which laid a foundation for deep research on the molecular mechanism of light on chicken reproductive performance.【Method】A total of 21610-week-old Guizhou yellow chickens with similar weight were randomly divided into 2 groups.One group had been given natural light group,and another had been given 1 more hour of lighting in the morning and evening.4 hens with 140-day-old,4 hens with 200-day-old and 4 hens with 480-day-old were slaughtered,and the tissue of pituitary,hypothalami,retina,tubal and ovarian were collected,so as to detect the differential expression of ONECUT1 gene through qRT-PCR.【Result】ONECUT1 gene had the most expression in muscle tissue,followed by fallopian tubes and ovaries,and the lowest in liver.In pituitary,hypothalamic,and ovarian tissues,the expression of ONECUT1 gene at peak egg production period(200 days of age)was significantly higher than those at the start date(140 days of age)and late egg laying period(480 days of age).The 200-day-old light enhancement group in pituitary tissue was significantly higher than that in the natural light group,and the natural light group at 140 days old in the fallopian tubes was significantly lower than the light enhancement group.【Conclusion】The expressions of ONECUT1 gene was exhibiting temporal and spatial expression patterns and photoperiod regulated,which was related to laying performance.
