摘  要:目的:通过网络药理学和免疫浸润探讨中药荜茇抗血吸虫病相关肝细胞癌的作用机制。方法:通过TCMSP、TCMIP数据库查找荜茇主要化学成分及其作用靶点;通过GeneCards、OMIM、TTD数据库收集血吸虫病及肝细胞癌相关靶点,构建“药物-成分-靶点”和“成分-靶点-通路”网络图,对药物和疾病的交集靶点进行蛋白质-蛋白质相互作用(PPI)分析;通过R语言对交集靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析;通过GEPIA和UALCAN数据库对潜在靶点进行表达差异及生存预后分析;通过TIMER数据库对核心靶点进行免疫浸润分析。结果:获得荜茇抗血吸虫病及肝细胞癌潜在靶点11个,其中肿瘤坏死因子(TNF)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、环加氧酶2(PTGS2)等可能是荜茇发挥作用的潜在靶点;KEGG通路富集主要涉及肿瘤坏死因子(TNF)信号通路、C型凝集素受体信号通路等,生物信息学结果表明葡萄糖-6-磷酸脱氢酶(G6PD)可能在荜茇治疗血吸虫病相关肝细胞癌过程中发挥重要作用。结论:本研究初步预测了荜茇抗血吸虫病相关肝细胞癌的分子机制,为进一步深入研究及临床应用提供了理论依据。Objective:To investigate the mechanism of action of Fructus Piperis Longi in the treatment of schistosomiasis-related hepatocellular carcinoma based on network pharmacology and immune infiltration.Methods:TCMSP and TCMIP databases were used to obtain the main chemical components of Fructus Piperis Longi and their action targets,and GeneCards,OMIM,and TTD databases were used to collect the targets associated with schistosomiasis and hepatocellular carcinoma;a drug-component-target network and a component-target-pathway network were constructed,and a protein-protein interaction analysis was performed for the intersecting targets of the drug and the disease.R language was used to perform the gene ontology functional enrichment analysis and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis of the intersecting targets.GEPIA and UALCAN databases were used to perform the differential expression and survival/prognosis analyses of potential targets,and TIMER database was used to perform an immune infiltration analysis of core targets.Results:A total of 11 potential targets were obtained for Fructus Piperis Longi in the treatment of schistosomiasis-related hepatocellular carcinoma,among which tumor necrosis factor(TNF),interleukin-1β,interleukin-6,interleukin-10,and prostaglandin endoperoxide synthase 2(PTGS2)might be the potential action targets of Fructus Piperis Longi.The KEGG pathway enrichment analysis mainly involved the TNF signaling pathway and the C-type lectin receptor signaling pathway,and the bioinformatics analysis showed that glucose-6-phosphate dehydrogenase might play an important role in the treatment of schistosomiasis-related hepatocellular carcinoma by Fructus Piperis Longi.Conclusion:This article preliminarily predicts the molecular mechanism of Fructus Piperis Longi in the treatment of schistosomiasisrelated hepatocellular carcinoma,which provides a theoretical basis for further in-depth research and clinical application.
