摘 要:目的:探讨柚皮苷(Nar)是否通过调节氧化应激及凋亡对糖尿病大鼠心肌组织缺血再灌注损伤(IRI)发挥保护作用。方法:SD大鼠24只,随机分为对照组(control组)、糖尿病缺血再灌注损伤组(D-IRI组)、柚皮苷低剂量+糖尿病缺血再灌注损伤组(L-Nar+D-IRI组)、柚皮苷高剂量+糖尿病缺血再灌注损伤组(H-Nar+D-IRI组),每组6只。应用超氧化物歧化酶(SOD)和丙二醛(MDA)试剂盒检测4组大鼠心肌组织中SOD和MDA水平。苏木精-伊红(HE)染色法观察心肌组织病理变化,氯化三苯基四氮唑(TTC)检测心肌梗死面积,应用原位末端标记测定法(TUNEL)检测大鼠心肌细胞凋亡变化,蛋白质免疫印迹法(Western Blot)检测B细胞淋巴瘤/白血病2(Bcl-2)、Bax蛋白表达量。结果:L-Nar+D-IRI组心肌组织SOD、Bcl-2表达高于D-IRI组(P<0.05);H-Nar+D-IRI组SOD、Bcl-2表达高于L-Nar+D-IRI组(P<0.05)。H-Nar+D-IRI组心肌MDA、凋亡率、Bax表达低于L-Nar+D-IRI组(P<0.05);L-Nar+D-IRI组心肌组织损伤及梗死面积轻于D-IRI组。H-Nar+D-IRI组心肌组织损伤程度及梗死面积轻于L-Nar+D-IRI组。结论:柚皮苷通过抑制心肌氧化应激和凋亡,对糖尿病SD大鼠心肌缺血再灌注损伤发挥保护作用。Objective:To investigate the protective effect of naringin on myocardial ischemia-reperfusion injury in diabetic rats by regulating oxidative stress and apoptosis.Methods:Twenty-four rats were randomly divided into control group(control),diabetic ischemia-reperfusion injury group(D-IRI),naringin low-dose+diabetic ischemia-reperfusion group(L-Nar+D-IRI),and naringin high-dose+diabetic ischemia-reperfusion group(H-Nar+D-IRI),with 6 rats in each group.The levels of superoxide dismutase(SOD)and malonaldehyde(MDA)in myocardium of rats in four groups were detected by SOD and MDA kits.Hematoxylin-eosin staining method was used to observe the pathological changes of myocardial tissue,and triphenyltetrazole chloride(TTC)was used to detect the size of myocardial infarction.Terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL)assay was used to detect the apoptosis of rat myocardial tissue,and Western Blot was used to detect the protein expression levels of Bcl-2 and Bax.Results:The expression of SOD and Bcl-2 in L-Nar+D-IRI group was higher than that in D-IRI group(P<0.05).The expression of SOD and Bcl-2 in H-Nar+D-IRI group was higher than that in L-Nar+D-IRI group(P<0.05).The MDA level,apoptosis rate,and Bax expression in H-Nar+D-IRI group were lower than those in L-Nar+D-IRI group(P<0.05).The injury degree and infarct size of myocardial tissue in L-Nar+D-IRI group were lighter than those in D-IRI group.The myocardial injury and infarct size in H-Nar+D-IRI group were lower than that in L-Nar+D-IRI group.Conclusion:Naringin plays some protective role in myocardial ischemia reperfusion injury in diabetic rats by inhibiting myocardial oxidative stress and apoptosis.
