摘  要:为探究氮离子注入对药用植物金莲花(Trollius chinensis)生理特性的影响,本研究以金莲花种子为试验材料,利用离子注入诱变技术把不同剂量的氮离子注入金莲花种子,测定了金莲花的发芽率、可溶性蛋白含量、叶绿素含量、丙二醛(MDA)含量、过氧化氢(H_(2)O_(2))含量、抗氧化酶活性和抗氧化酶相关基因表达量。结果表明,随着注入剂量的增加,金莲花种子的发芽率、叶绿素含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的活性均呈现“升-降-升-降”的“马鞍型”变化趋势,过氧化物酶(POD)活性呈现“升-降-升”的变化趋势。较低剂量氮离子注入时,金莲花MDA含量和H_(2)O_(2)含量低于较高剂量组。3.0×10^(15)ions·cm^(-2)的氮离子注入可上调金莲花幼苗抗氧化相关基因的表达。综合多项指标认为,较低剂量氮离子注入可提高金莲花种子的活力,促进幼苗的生长发育,增加幼苗抗氧化酶的活性,增强幼苗的抗逆性,其诱变的优选剂量为3.0×1015ions·cm^(-2)。本研究结果为进一步研究金莲花氮离子注入诱变育种提供了理论依据。In order to explore the effects of nitrogen ion implantation on the physiological characteristics of Trollius chinensis.Different doses of nitrogen ions were implanted into the seeds of Trollius chinensis.The effects of nitrogen ion implantation on the germination rate,soluble protein content,chlorophyll content,malondialdehyde(MDA)content,hydrogen peroxide(H2O2)content,antioxidant enzyme activity,and expressions of antioxidant enzyme related genes in Trollius chinensis were studied.The results showed that,with the increase of implantation dose,the germination rate,chlorophyll content,the activities of superoxide dismutase(SOD)and catalase(CAT)show a“saddle-shaped”trend of“up-down-up-down”,while the activity of peroxidase(POD)shows a trend of“up-down-up”.The content of MDA and H2O2 in Trollius chinensis treated with the lower dose nitrogen ion implantation is lower than that treated with higher dose.Nitrogen ion implantation with 3.0×10^(15) ions·cm^(-2) can upregulate the expression of antioxidation-related genes in Trollius chinensis seedlings.This study suggests that relatively low doses of nitrogen ion can improve the seed vigor of Trollius chinensis,promote the growth of its seedling,increase the activity of antioxidant enzymes in its seedlings and enhance the stress resistance of its seedlings.The optimal dose for nitrogen ion implantation mutagenesis of Trollius chinensis is 3.0×10^(15) ions·cm^(-2).The results can provide a theoretical basis for further research on mutation breeding of Trollius chinensis by nitrogen ion implantation.
