摘  要:针对辣椒缺乏稳定且高多态性的SNP标记问题,根据辣椒高密度遗传图谱和物理图谱的对应关系,以20 cM间距设计KASP标记,共设计了234对KASP引物。利用F2分离群体筛选获得具有多态性的引物159对,标记设计成功率67.9%。利用来自14个引种地的207份材料构成的辣椒群体,对引物进行了高通量体系转化和标记的多态性、稳定性验证。通过高通量基因分型体系对挑选出的141个标记进行多态性检测,其中130个KASP标记成功分型,成功率为92%,且在辣椒材料中具有多态性的标记数量变化范围为24~129个,对应的多态性位点百分率范围为18.46%~99.23%。试验开发了一组包含130个辣椒KASP标记,均匀分布在辣椒的12条染色体上,通过对国内的14个引种来源地辣椒品种的分型,证明了这套标记多态性高、普适性强,适用于辣椒品种的高通量基因分型。Aiming at the lack of stable and highly polymorphic SNP markers of peppers,KASP markers were designed at a spacing of 20 cM according to the correspondence between the high-density genetic map and the physical map of peppers,and a total of 234 pairs of KASP primers were designed.F2 isolated population screening was used to obtain 159 pairs of primers with polymorphism,and the success rate of labeling design was 67.9%.Using a group of 207 peppers from 14 introduction sites,the polymorphism and stability of primers were verified for high-throughput system transformation and labeling.The polymorphism of 141 selected markers was detected by high-throughput genotyping system,of which 130 KASP markers were successfully typed,with a success rate of 92%,and the number of markers with polymorphism in pepper materials varied from 24-129,and the corresponding polymorphism site percentage range was 18.46%-99.23%.A set of KASP markers containing 130 pepper KASP markers was developed and evenly distributed on the 12 chromosomes of peppers,and through the typing of 14 domestic pepper varieties from the source of introduction,it was proved that this set of marker polymorphisms is highly polymorphic and universal,and is suitable for high-throughput genotyping of pepper varieties.
