摘 要:目的探讨蛋白激酶D(PKD)在单核细胞增生李斯特菌(Lm)感染Caco-2细胞中的作用及其机制。方法建立Lm感染Caco-2细胞模型,qRT-PCR检测PKD1、PKD2和PKD3 mRNA表达水平。加入PKD抑制剂CID755673后,Lm感染Caco-2细胞,CCK-8法测定细胞存活率,Western blot法检测细胞炎症相关蛋白和ERK1/2信号通路蛋白的表达。结果Lm感染促进Caco-2细胞表达PKD;与Lm组相比,抑制PKD可明显降低Caco-2细胞存活率(F=4.831,P<0.05),明显抑制炎症相关蛋白NLRP3、p-IκBα(F_(NLRP3)=35.02,F_(p-IκBα)=45.19,均P<0.01)和ERK1/2蛋白(F=326.9,P<0.001)的表达。结论PKD-ERK1/2信号通路在Lm感染Caco-2细胞中发挥重要调控作用。This study was aimed at investigating the role and mechanism of protein kinase D(PKD)in the infection of Caco-2 cells by Listeria monocytogenes(Lm).The mRNA expression levels of PKD1,PKD2,and PKD3 were determined by qRT-PCR in Caco-2 cells infected with Lm.After the addition of the PKD inhibitor CID755673,Caco-2 cells were infected with Lm;cell viability was determined with CCK-8 assays;and the expression of inflammation related proteins and ERK1/2 signaling pathway proteins was determined by western blotting.The expression of PKD was elevated in Caco-2 cells infected with Lm.Compared with the Lm group,the group with inhibition of PKD showed significantly lower Caco-2 cell viability(F=4.831,P<0.05),as well as significant inhibition of the expression of the inflammation related proteins NLRP3 and p-IκBα(F_(NLRP3)=35.02,F_(p-IκBα)=45.19,all P<0.01)and the proteins ERK1/2(F=326.9,P<0.001).These results indicated that the PKD-ERK1/2 signaling pathway played an important regulatory role in Caco-2 cells infected with Lm.
