摘 要:棉花黄萎病严重影响中国的棉花产量和品质。本研究以海岛棉cDNA为模板扩增得到3个DAHP基因片段,分别命名为GbDAHP-1、GbDAHP-6、GbDAHP-7。序列分析显示,GbDAHP-1基因全长为1755 bp,编码539个氨基酸,理论分子量为59.4 kD,等电点为8.70;GbDAHP-6基因全长为1644 bp,编码519个氨基酸,理论分子量为57.17611 kD,等电点为8.94;GbDAHP-7基因全长为1554 bp,编码517个氨基酸,理论分子量约为57.17714 kD,等电点为8.12。通过在线软件分析得知3个蛋白质的理化性质相似,均为亲水性蛋白,不含跨膜运输结构域,位于叶绿体。系统发育树显示:GbDAHP-1、GbDAHP-6和GbDAHP-7基因与陆地棉、雷蒙德氏棉、可可的序列相似度较高,与陆地棉以及雷蒙德氏棉的蛋白质相似度最高,亲缘关系最近。在黄萎病菌诱导下GbDAHP基因显示先上升后下降的趋势,表明该基因受到黄萎病菌诱导表达。Verticillium wilt of cotton has seriously affected the yield and quality of cotton in China.In this study,three DAHP gene fragments named GbDAHP-1,GbDAHP-6 and GbDAHP-7 were amplified from Gossypium barbadense cDNA.Sequence analysis showed that GbDAHP-1 gene was 1755 bp,it encodes 539 amino acids,the theoretical molecular weight is 59.4 kD and the isoelectric point is 8.70;GbDAHP-6 gene was 1644 bp,it encodes519 amino acids,the theoretical molecular weight is about 57.17611 kD and the isoelectric point is 8.94;GbDAHP-7 gene was 1554 bp,it encodes 517 amino acids,the theoretical molecular weight is about 57.17714 kD and the isoelectric point is 8.12.Through online software analysis,we found that the physical and chemical properties of the three proteins were similar,they were hydrophilic proteins,without transmembrane transport domain,located in chloroplast.Phylogenetic tree showed that GbDAHP-1,GbDAHP-6 and GbDAHP-7 genes had high sequence similarity with Gossypium hirsutum,Gossypium raimondii and Theobroma cacao,and had the highest protein similarity with Gossypium hirsutum and Gossypium raimondii.Under the induction of Verticillium dahliae,GbDAHP gene showed an upward trend at first and then a downward trend,indicating that GbDAHP gene was induced by Verticillium dahliae.
