摘  要:为了探究PAO基因在荔枝(Litchi chinensis Sonn.)生长发育过程中的生物学功能,本研究基于荔枝全基因组数据,利用生物信息学方法筛选与鉴定荔枝PAO基因(LcPAO)成员,并对其理化性质、系统发育、结构、染色体定位、顺式作用元件、蛋白结构、miRNA靶位点和表达模式进行了分析。结果表明,一共鉴定得到10个LcPAO基因,全部为亲水蛋白。系统发育分析发现LcPAO落在4个亚族中的Ⅰ、Ⅱ和Ⅲ亚族。LcPAO1~LcPAO3和LcPAO9~LcPAO10属于典型的PAO蛋白,LcPAO4~LcPAO8还含有一个特异的SWIRM结构域,属于非典型PAO蛋白。顺式作用元件分析表明其主要含光响应元件、逆境胁迫相关元件以及激素响应相关元件。miRNA靶位点分析发现LcPAO基因受到22个miRNAs的靶向调控。转录组表达模式分析发现,除了LcPAO1和LcPAO4,余下的LcPAO基因可能与荔枝果皮着色相关。所有的LcPAO基因均可能与荔枝的化学控花相关。除了LcPAO1所有的LcPAO基因均可能与荔枝果实脱落相关。本研究为深入研究LcPAO基因的功能和调控机制提供参考。In order to explore the biological function of polyamine oxidase(PAO)gene in the growth and development of Litchi chinensis,based on the whole genome data of Litchi,the PAO gene members of Litchi chinensis(LcPAO)were screened and identified,and their physicochemical properties,structure,phylogeny,chromosomal localization,cis-acting elements,protein structure,miRNA target site and expression patterns were analyzed by bioinformatics.The results showed that a total of 10 LcPAO genes were identified and all of them were hydrophilic proteins.Phylogenetic analysis exhibited that LcPAO genes fell intoⅠ,ⅡandⅢsubgroups of the four subgroups.LcPAO1~LcPAO3 and Lc PAO9~LcPAO10 belonged to typical PAO proteins,and LcPAO4~LcPAO8belonged to atypical PAO proteins which contained a specific SWIRM domain.Cis-acting elements analysis displayed that them mainly contained light response elements,stress related elements and hormone response related elements.miRNA target site analysis found that LcPAO genes were targeted by 22 miRNAs.Transcriptome expression pattern analysis demonstrated that in addition to LcPAO1 and LcPAO4,the remaining LcPAO genes may be related to Litchi chinensis pericarp coloring.All LcPAO genes may be involved in the chemical flower control of Litchi.All LcPAO genes except LcPAO1 may be related to Litchi fruit abscission.This study provides a reference for further study on the function and regulation mechanism of LcPAO genes.
