摘 要:本研究评估了水花生植物提取物绿色合成纳米氧化锌的可行性并评价了其对猪繁殖与呼吸综合征病毒(PRRSV)的体外抗病毒效果。采用紫外-可见光谱、扫描电镜观察以及能谱扫描仪分析等多种方法交互验证纳米氧化锌颗粒的形成,并采用MTT比色法在预防、阻断、直接灭活三种药物作用方式下对其体外抗PRRSV的活性进行评估。结果表明纳米氧化锌颗粒在反应液中呈胶体状态,最大吸收峰在波长260 nm处;晶体颗粒呈六棱柱形,其粒径为34.8~314.2 nm,平均尺寸为43.8 nm。纳米氧化锌药物细胞安全质量浓度为0.125 mg/mL。纳米氧化锌针对PRRSV在预防、阻断以及直接杀灭三种作用方式下对细胞的最高平均保护率分别为35.1%、63.7%、73.9%。结果提示,纳米氧化锌在开发作为抗PRRSV的消毒剂以及阻断剂方面很有应用前景。该研究说明利用水花生植物提取液可以合成纳米氧化锌颗粒,而且显示了良好的体外抗病毒效果,这也为今后新兽药研发以及入侵植物资源化利用提供了借鉴。In order to assess the feasibility of green synthesis of zinc oxide nanoparticles(ZnO-NPs),the plant extract of Alternanthera philoxeroides was used and its antiviral effects against porcine reproductive and respiratory syndrome virus(PRRSV) were evaluated.The formation of ZnO-NPs was cross-validated via UV-Visible spectroscopy,scanning electron microscopy(SEM) and energy-dispersive X-ray(EDX) analysis.The in vitro antiviral activity of ZnO-NPs against PRRSV was evaluated with MTT assay in three models of drug action(e.g.,preventive effect,blocking effect and direct inactivate).The Results showed that the synthesized ZnO-NPs formed colloids in the reaction solution.And a maximum absorption peak at 260 nm was observed.The crystal structure of ZnO-NPs was in the shape of hexagonal prism(size range:34.7-314 nm,43.8 nm in average).The safe mass concentration of ZnO-NPs was 0.125 mg/mL.The highest average protection rates of ZnO-NPs against PRRSV were 35.1%,63.7% and 73.9%,respectively for prevention,blocking and direct inactivation.The results suggested that ZnO-NPs showed great promise in application as disinfectant or blocking agent against PPRSV.All the results revealed the feasibility of synthesizing ZnO-NPs with plant extract of Alternanthera philoxeroides and exhibited good antiviral effects against PRRSV in vitro,which provides a reference for development of new veterinary pharmaceuticals and resourceful utilization of invasive plants in the future.
