摘 要:目的:探讨红景天苷(salidroside,SAL)对不同组织离体血管张力的药理作用及引起最大舒缩的浓度。方法:雄性SD大鼠处死后,取出组织并放入4℃PSS溶液中,在显微镜下分离动脉,剪出2 mm的血管环备用。采用给药前后对照的方法,分为对照组(SAL:0μg·mL^(-1))和实验组(0.3-1861μg·mL^(-1)),用微血管张力测定法测定血管张力。结果:(1)SAL对不同组织器官离体微血管张力的影响有选择性作用。给药后,大脑中动脉、基底动脉、颈总动脉,肺动脉、冠状动脉、胸主动脉、腹主动脉、肠系膜动脉、股动脉的IC_(50)值分别为1241.4,1230.3,11498.7,1230.3,125.7,1488.9,759.3,1491,1227μg·mL^(-1)。与对照组相比,差异均有统计学意义(P<0.05)。(2)SAL舒张各血管的浓度为:脑中动脉(961~1261μg·mL^(-1))、基底动脉(961~1861μg·mL^(-1))、肺动脉(0.3~2336μg·mL^(-1))、冠状动脉(0.3~1111μg·mL^(-1))、胸主动脉(961~2261μg·mL^(-1))、腹主动脉(6~2011μg·mL^(-1))、肠系膜动脉(961~2011μg·mL^(-1))、股动脉(480~2011μg·mL^(-1))。结论:SAL对不同组织器官离体微血管张力的影响有选择性作用,为红景天苷用于防治不同组织器官缺血性疾病提供实验数据。Objective:To investigate the pharmacological effect of salidroside (SAL) on vascular tension in different tissues in vitro and the concentration of salidroside (SAL) causing maximum contraction.Method:After the male SD rat was sacrificed,the tissue was removed and put into 4℃ PSS solution.The artery was separated under the microscope,and a 2 mm vascular ring was cut out for later use.The pre-and post-administration control method was used and divided into a control group (SAL:0μg·mL^(-1)) and experimental group (0.3~1861μg·mL^(-1)).Vascular tone was detected by microvascular tone assay.Result:(1)Salidroside had a selective effect on microvascular tension of different tissues and organs in SD rats.After administration,the IC_(50)values of middle cerebral artery,basilar artery,common carotid artery,pulmonary artery,coronary artery,thoracic aorta,abdominal aorta,mesenteric artery and femoral artery were 1241.4,1230.3,11498.7,1230.3,125.7,1488.9,759.3,1491,and 1227μg·mL^(-1),respectively.Compared with the control group (0μg·mL^(-1) SAL),the differences were statistically significant (P<0.05).(2) The dose of salidroside for vasodilation in different tissues and organs was:cerebral artery (961~1261μg·mL^(-1)),basilar artery (961~1861μg·mL^(-1)),pulmonary artery (0.3~2336μg·mL^(-1)),coronary artery (0.3~1111μg·mL^(-1)),thoracic aorta (961~2261μg·mL^(-1)),abdominal aorta (6~2011μg·mL^(-1)),mesenteric artery (961~2011μg·mL^(-1)),femoral artery (480~2011μg·mL^(-1)).Conclusion:Salidroside has a selective effect on microvascular tension in different tissues and organs,which provides experimental data for preventing and treating ischemic diseases in different tissues and organs.
