摘 要:目的:Aβ25-35淀粉酶注射SD大鼠建立老年痴呆大鼠(AD)模型,评价还神至圣汤对AD大鼠学习记忆能力的影响。方法:50只SD大鼠随机分为正常对照组,模型组、实验药物10.2、20.4、40.8 g.kg^(-1)剂量组,10只/组。除正常对照组外,其余各组大鼠海马区注射Aβ25-35淀粉酶建立AD模型。建模后第2天各组动物每日给药,连续30天,正常对照组、模型组给予等体积生理盐水,末次给药后24 h,剖取大鼠右侧海马组织,采用WB法测定海马区PI3K、AKT、P-AKT、GSK3β、P-GSK3β蛋白水平。模型建立后14天,开展Morris水迷宫实验测定AD大鼠学习记忆能力。结果:还神至圣汤各剂量组明显减少平台逃避潜伏期,提高穿越平台次数和目标象限停留时间百分比(P<0.05)。还神至圣汤20.4、40.8 g.kg^(-1)剂量组PI3K、p-Akt、p-GSK3β蛋白含量有明显增加(P<0.05,40.8 g.kg^(-1)剂量组p-GSK3β除外)。P-Akt/Akt、P-GSK3β/GSK3β比值也明显增加(P<0.05)。结论:还神至圣汤可改善由Aβ25-35淀粉样蛋白诱导的AD模型大鼠的学习记忆能力,其作用可能通过调控PI3K/AKT/GSK3β通过信号通路实现的。Objective:The rat model of senile dementia (AD) was established by injecting Aβ25-35 amylase in order to explore the effects of Huanshenzhisheng Decoction(HSZS) on the learning and memory ability of AD rats.Method:Fifty health rats were randomly divided into five groups as the control group(NS),the model group,three dosages of HSZS groups (10.2g.kg^(-1),20.4 g.kg^(-1),40.8 g.kg^(-1)),ten rats in each group.AD model was established by injecting Aβ25-35 amylase into hippocampus of rats in each group,except control group.In the 2nd day,rats in the control and model groups were treated with saline,while rats in HSZS groups were orally treated with different doses of HSZS for thirty days,Morris water maze test was conducted to determine the learning and memory ability of AD rats in 14th days after the model establishment.Twenty-four hours after the last administration,the right hippocampal tissue of rats was dissected,and the protein levels of PI3K,AKT,p-Akt,GSK3β and p-GSK3βin the hippocampus were detected by WB method.Result:The platform escape latency of rats in the model control group was decreased,and the times of crossing the platform and the percentage of target quadrant residence time were significantly increased in each group of HSZS (p<0.05).The level of PI3K,p-Akt,p-GSK3β was increased in group of middle dosage,as the same as PI3K,p-Akt in high dosage.The ratio of P-Akt/Akt and P-GSK3β/GSK3β were significantly increased (P<0.05).Conclusion:HSZS Decoction can improve the learning and memory ability of AD rats induced by Aβ25-35 amyloid protein,which may be achieved by regulating PI3K/AKT/GSK3β signaling pathway.
