摘 要:目的优选经典名方三石汤的提取工艺。方法在参考古籍中三石汤制备工艺的基础上,以绿原酸、苦杏仁苷含量和出膏率作评价指标进行正交试验;采用HPLC法建立三石汤指纹图谱,确定共有峰并进行相似度评价;结合正交实验结果优化三石汤提取工艺参数并验证。结果三石汤HPLC指纹图谱确认了7个共有峰,并确定其中2个化合物,10批三石汤样品相似度均大于0.95;根据综合评分与指纹图谱相似度结果得到三石汤最适宜提取工艺为10倍加水量,浸泡30 min,提取2次,每次20 min。结论优选后的工艺提取率高、稳定可行,以期为经典名方三石汤复方制剂的开发研究奠定基础。Objective To optimize the extraction process of compound Sanshi Decoction.Methods On the basis of reference to the preparation technology of Sanshi Decoction in ancient books,Orthogonal test design was the contents of chlorogenic acid,a⁃mygdalin and dry extract yield in Sanshi Decoction were used as evaluation indexes.The fingerprint of Sanshi Decoction was es⁃tablished by HPLC,determine the common peak and evaluate the fingerprint similarity.The extraction parameters of compound Sanshi Decoction were optimized according to the orthogonal experimental results and the experimental results were verified.Re⁃sults The HPLC fingerprint of Sanshi Decoction showed 7 common peaks,and 2 compounds were identified.The similarity of 10 batches of Sanshi Decoction samples were all more than 0.95.According to comprehensive score and the results of similarity of fingerprint,the optimum extraction process of compound Sanshi Decoction was 10 times the amount of water,soaking for 30 mi⁃nutes,extracted twice,20 minutes each time.Conclusion The extraction rate of the optimized process is high,stable and feasi⁃ble,which lays a foundation for the study of the quality standard of Sanshi Decoction compound preparation.
