摘  要:目的:观察补阳还五汤对大鼠脑缺血再灌注损伤后脑组织铁转运相关蛋白表达及铁死亡的影响。方法:32只雄性SD大鼠随机分为假手术组、模型组、依达拉奉组和补阳还五汤组,每组8只。除假手术组外,其他组均建立大鼠大脑中动脉阻塞(MCAO)模型。除假手术组外,其余3组均于术前7 d和再灌注后连续3 d分别给予等量0.9%氯化钠溶液、依达拉奉腹腔注射,补阳还五汤颗粒剂灌胃,每日1次。采用TTC染色检测各组大鼠脑梗死面积,HE染色和尼氏染色观察缺血侧海马CA3区病理组织形态变化,免疫组织化学法观察缺血侧海马CA3区Fn、Tf、TfR、FPN1蛋白表达情况,Western Blot法检测铁死亡相关蛋白Nrf2、HO-1、GPX4蛋白表达情况。结果:与模型组比较,补阳还五汤组脑梗死面积显著减小(P<0.01),尼氏小体计数显著增多(P<0.05),病理损伤减轻,Tf、TfR、Fn蛋白表达显著降低(P<0.05),FPN1、Nrf2、HO-1和GPX4蛋白表达显著升高(P<0.05)。结论:补阳还五汤通过调节脑缺血再灌注损伤大鼠缺血侧海马CA3区铁转运蛋白的表达,调控Nrf2/HO-1/GPX4信号通路,减轻脑缺血损伤。Objective:To observe the effect of Buyang Huanwu Decoction on the expression of iron transport-related proteins and ferroptosis after cerebral ischemia reperfusion injury in rats.Methods:A total of 32 SD male rats were randomly divided into sham group,model group,edaravone group and Buyang Huanwu Decoction group,8 rats in each group.Except for sham group,the rat models of middle cerebral artery occlusion(MCAO)were established in other groups.Except for sham group,another three groups were given an equal amount of 0.9%sodium chloride solution and edaravone by intraperitoneal injection,Buyang Huanwu Decoction granules by gavage,once a day for 7 days before the operation and for 3 consecutive days after the reperfusion,respectively.TTC staining was used to detect the cerebral infarct size in each group,HE staining and Nissl staining were used to observe the pathological histomorphological changes of hippocampal CA3 region on the ischemic side,immunohistochemical method was used to detect the protein expression of Fn,Tf,TfR and FPN1 in hippocampal CA3 region,and Western Blot was used to detect the protein expression of ferroptosis related protein Nrf2,HO-1 and GPX4.Results:Compared with the model group,the cerebral infarct size in Buyang Huanwu Decoction group was significantly reduced(P<0.01),the numbers of Nissl body significantly increased(P<0.05),the pathological damage was alleviated,the protein expression of Tf,TfR and Fn were significantly reduced(P<0.05),and the protein expression of FPN1,Nrf2,HO-1 and GPX4 were significantly increased(P<0.05).Conclusion:Buyang Huanwu Decoction can regulate the expression of iron transporter in hippocampal CA3 region of rats with cerebral ischemia-reperfusion injury,regulate Nrf2/HO-1/GPX4 signal pathway,and reduce cerebral ischemic injury.
