摘 要:目的:探讨仙苓莲夏方对三阴性乳腺癌小鼠的治疗作用及对肠道菌群和免疫功能的影响。方法:21只雌性BALB/c小鼠随机分为正常组、模型组和仙苓莲夏方组,每组7只,连续给药4周。治疗结束后剥离肿瘤,计算体积;采集外周血进行血常规检测,流式细胞术分析血液和骨髓中的免疫细胞;无菌条件下收集各组小鼠粪便,提取肠道菌群DNA,对V3-V4区进行双端测序及生物信息学分析。结果:与模型组比较,仙苓莲夏方显著减小肿瘤重量、体积及脾脏重量,降低外周血白细胞、血小板、单核细胞、淋巴细胞和中性粒细胞的数量,升高骨髓中短期造血干细胞(ST-HSC)和巨核系-红系祖细胞(MEP)及外周血中自然杀伤细胞(NK)的百分比,同时降低外周血中Treg、M-MDSC的百分比(P<0.05,P<0.01)。拟杆菌门、厚壁菌门和变形菌门是各组小鼠的主要菌群,且变形菌门的相对丰度在模型组中显著降低,仙苓莲夏方干预后显著升高(P<0.05)。结论:仙苓莲夏方可以有效抑制三阴性乳腺癌小鼠肿瘤生长,保护骨髓造血,降低外周血中抑制性细胞、升高NK细胞的百分比,调节肠道菌群紊乱。Objective:To investigate the therapeutic effect of Xianling Lianxia Formula on triple negative breast cancer mice and its influence on intestinal flora and immune function.Methods:Twenty-one female BALB/c mice were randomly divided into normal group,model group and Xianling Lianxia Formula(XLLX)group,with 7 mice in each group,for 4 consecutive weeks.After treatment,the tumor was dissected and its volume was calculated,peripheral blood was collected for routine blood testing,and flow cytometry was used to analyze immune cells in blood and bone marrow,feces of mice in each group were collected,and intestinal flora DNA was extracted.The V3 to V4 regions were double-ended sequenced and analyzed for trustworthiness.Results:Compared with the model group,XLLX significantly reduced the weight,volume of the tumor and spleen weight,decreased the number of white blood cells,platelets,monocytes,lymphocytes and neutrophils in peripheral blood,increased the percentage of short-term HSCs(ST-HSCs)and megakaryocyte-erythrocyte progenitors(MEP)in the bone marrow and natural killer cells(NK)in the peripheral blood,and reduced the percentage of Treg and M-MDSC cells in peripheral blood(P<0.05,P<0.01).Bacteroidetes,Firmicutes and Proteobacteria were the main flora in each group of mice,and the relative abundance of Proteobacteria decreased in the model group,and increased significantly after the intervention of XLLX(P<0.05).Conclusion:XLLX can effectively inhibit tumor growth in triple negative breast cancer mice,protect bone marrow hematopoiesis,reduce suppressor cells in peripheral blood,increase the percentage of NK cells,and regulate intestinal flora disturbance.
