摘  要:为有效降低纤维乙醇生产成本,本研究以玉米秸秆作为原料,烟曲霉作为产酶及酶解菌株,采用嗜鞣管囊酵母以半同步糖化发酵方法生产乙醇,对烟曲霉酶解过程、嗜鞣管囊酵母发酵生产乙醇的酶解和发酵条件分别进行了优化。结果表明,在10 g玉米秸秆中加入90 m L 0.2 mol/L Na OH溶液,121℃灭菌20 min并冷却到室温后用5 mol/L无菌H2SO4或10 mol/L Na OH调节p H至4.5,加入无菌饱和硫酸铵至终浓度为10 g·L^(-1),该组成的发酵培养基中接入8%烟曲霉种子,酶解12 h后接入4%酵母种子,酶解发酵共计28 h,玉米秸秆降解率可达94.2%,获得乙醇31.8 g·L^(-1)。这说明预处理后无需固液分离及脱毒,采用烟曲霉酶解稀碱预处理玉米秸秆,以及嗜鞣管囊酵母发酵能经济有效低成本获得生物乙醇。In order to effectively reduce the cost of cellulosic ethanol production,the corn straw was used as the raw material,the As⁃pergillus fumigatus was used as the enzyme producing and enzymolysis strain,and a semi-simultaneous saccharification and fermentation method was adopted to produce the ethanol with Saccharomyces tannophilus.The enzymatic hydrolysis of Aspergillus fumigatus and fermentation conditions of ethanol production by Saccharomyces tannophilus were optimized.The results showed that when the fermentation medium composed of 10 g corn straw with 90 mL of 0.2 mol/L NaOH solution,sterilized at 121℃for 20 min and cooled to room temperature,adjusted to pH 4.5 with 5 mol/L sterile H2SO4 or 10 mol/L NaOH,and sterile saturated ammonium sulfate was added to its final concentration of 10 g·L^(-1) was supplemented with 8%of Aspergillus fumigatus innoculum,then 4%yeast innoculm was added in after 12 h of enzymolysis,the corn straw degradation rate was 94.2%,and an ethanol concentration of 31.8 g·L^(-1) was obtained in a total enzymolysis and fermentation time of 28 h.It was indicated that with no requirements for solid-liquid separation and detoxification after pretreatment,the Saccharomyces tannophilus can economically and effectively obtain the bioethanol by pretreating corn stover with enzymatic dilution of alkali by Aspergillus fumigatus.
