摘　　要：肽池(peptide pool)是一系列特定长度氨基酸的排列组合,通常可以模拟体外抗原激活机体免疫反应,产生针对目标病原体的特异性抗体,成为研究新发和再发传染病的有效工具。肽池分为生物肽池、化学合成肽池和噬菌体展示肽池,不同种类的肽池制备方式不同且各有优缺点。近年来,化学合成肽池不断发展完善,与酶联免疫斑点试验、流式细胞检测技术结合,刺激免疫细胞产生可检测的目标因子,用于探索疫苗免疫途径、评价佐剂效果和筛选抗肿瘤单抗,可以提高科学实验效率,节约原料。现就抗原肽池制备技术及其在传染病病原抗原表位筛选和免疫效果评估中的应用作一概述。Peptide pool,a series of combinations of specific length of amino acids,has become an effective tool for the investigation of emerging and reemerging infectious diseases due to its function of simulating antigens to activate the body's immune response in vitro and producing specific antibodies against the target pathogen.It can be subdivided into biological peptide pool,chemical synthetic peptide pool and phage display peptide pool,featured by distinct preparation methods with their own advantages and disadvantages.In recent years,the combination of chemical synthetic peptide pool to enzyme-linked immunospot assay and flow cytometry technology has been continuously developed and improved,and has been used to explore the immune pathway of vaccines,evaluate the effect of adjuvants and screen anti-tumor mab.This combination,with the characteristic of stimulating immune cells to produce detectable target factors,has advantages to improve the efficiency of scientific experiments and save raw materials.In this paper,the preparation technology of antigen peptide pool and its applications in the screening of antigen epitope and the evaluation of immune effect on infectious diseases are reviewed.
