摘  要:目的:建立灵芝孢子油中甘油三酯类成分含量的高效液相色谱-蒸发光散射检测测定法。方法:采用ODS(100 mm×4.6 mm,2.7μm)色谱柱,以有机混合溶剂乙腈-无水乙醇-甲基叔丁基醚(45∶45∶10)为流动相等度洗脱,采用蒸发光散射检测法,分离了灵芝孢子油中8个主要甘油三酯类成分,以峰面积对数(lg A)为Y,质量浓度对数(lg C)为X进行线性回归并测定含量。结果:方法验证结果表明,各组分峰间的分离度符合要求,它们的蒸发光散射响应均在4.0~200μg·mL^(-1)浓度范围呈现良好的线性关系(r>0.999),平均加样回收率为96.63%~107.53%(RSD<5.0%)。结论:该方法专属性好,分析洗脱时间适宜,定量准确,满足灵芝孢子油中甘油三酯类成分质量控制的要求。Objective:To establish an HPLC-ELSD method for the assay of triglycerides contained in Ganoderma lucidum spore oil.Methods:Separation of 8 main triglycerides in Ganoderma lucidum spore oil was performed on an ODS(100 mm×4.6 mm,2.7μm)column by isocratic elution using acetonitrile-anhy-drous alcohol-methyl tertbutyl ether with the volume ratio of 45∶45∶10 as the mobile phase and evaporative light-scattering detection.They were quantified by a calibration curve method using linear regression algo-rithm of the log peak areas(lgA)as Y to log mass concentrations(lgC)as X.Results:The results of method validation showed that the resolution between the peaks of each component met the requirements,and all their evaporative light scattering responses showed good linear relationship in the mass concentra-tion range of 4.0-200μg·mL^(-1)(r>0.999).The average recoveries were between 96.63%and 107.53%(RSD<5.0%).Conclusion:This method has good specificity,suitable elution time and quantification accu-racy,which can be applied for quality control of triglycerides contents in Ganoderma lucidum spore oil.
