胶原诱导性关节炎大鼠盲肠内容物中短链脂肪酸分析

摘　　要：目的:建立气相色谱法检测大鼠盲肠内容物中短链脂肪酸的含量,分析正常大鼠和胶原诱导性关节炎大鼠盲肠内容物中短链脂肪酸含量变化。方法:使用1%甲酸水溶液提取盲肠内容物中短链脂肪酸,建立标准液标准曲线,并利用此方法检测正常大鼠和关节炎大鼠盲肠内容物中短链脂肪酸含量。结果:标准曲线显示出良好的线性关系,精密度、重复性、相对回收率以及稳定性均符合生物样品检测要求;与正常组相比,模型组的大鼠盲肠内容物中乙酸、丙酸和丁酸的含量显著降低,异戊酸的含量显著升高。结论:建立了高效稳定的检测大鼠盲肠内容物中短链脂肪酸的气相色谱法,发现正常大鼠和胶原诱导性关节炎大鼠盲肠内容物中短链脂肪酸含量存在差异。Objective:To establish a method for detecting the content of short-chain fatty acids(SCFAs)in cecum contents of rats by gas chromatography(GC),and the changes of cecal contents SCFAs in normal and collagen-induced arthritis rats were analyzed.Methods:1%formic acid water was used to extract SCFAs in cecum contents,the standard curve of standard solution was established and the content of SCFAs in cecum of normal and collagen-induced arthritis rats was detected by this method.Results:Standard curves showed good linear relationship.Precision,repeatability,relative recovery and stability all met the requirements of biological sample detection.Compared with the normal group,the content of acetic acid,propionic acid,and butyric acid in the cecum contents of the model group rats was significantly reduced,while the content of isovaleric acid was significantly increased.Conclusion:An efficient and stable gas chromatography method for detecting SCFAs in cecum contents of rats was established,and discovered differences in the content of SCFAs in the cecal contents between normal and collagen-induced arthritis rats.
