摘  要:目的 探究兔耳缘交替静脉滴注胺碘酮对血管损伤和修复的影响。方法 取90只大耳白兔为实验对象,随机分为A组、B组和C组,每组30只。A组在滴注完毕后立即采集标本,B组滴注完毕后72 h收集标本,C组滴注完毕后1周收集样本。再将每组进一步随机分为6个亚组,即1 h组、2 h组、3 h组、4 h组、5 h组、6 h组,每组5只,并分别每1 h、2 h、3 h、4 h、5 h、6 h更换到另一侧耳缘静脉通道给药。肉眼观察兔耳缘静脉炎程度;ELISA检测C反应蛋白(CRP)浓度;HE染色检测炎症程度;TUNEL染色检测血管内皮细胞凋亡水平。结果 A组中1~4 h组大耳白兔耳缘静脉均无明显肉眼可见的变化,5 h组1只大耳白兔输液部位红肿,6 h组1只大耳白兔出现血管轻微条索样改变。A组中1 h、2 h、3 h组CRP浓度比较差异无统计学意义(P>0.05);4 h、5 h、6 h组CRP浓度逐渐增加(P<0.05)。HE染色和TUNEL染色结果显示,A组4 h内血管炎症损伤程度最小,4 h开始炎症损伤逐渐增加,血管内皮细胞凋亡也明显增加;B组经72 h修复后血管炎症程度和细胞凋亡改善;C组经1周修复后血管炎症程度和细胞凋亡进一步改善。结论 交替静脉滴注胺碘酮在4 h内对静脉损伤的影响最小,兔耳缘血管内皮细胞的凋亡程度最轻,因此推荐最佳交替时间为4 h内。Objective To explore the effects of alternate intravenous instillation of amiodarone on the Vascular injury and repair of rabbit marrginal ear vein.Methods Ninety large-eared white rabbits were selected as experimental subjects and randomly divided into groups A,B and C,with 30 rabbits in each group.The samples of group A were collected immediately after the completion of the infusion,the samples of group B were collected 72 hours after the completion of the infusion,and the samples of group C were collected 1 week after the completion of the infusion.Each group was further randomly divided into 6 subgroups,namely,1-hour group,2-hour group,3-hour group,4-hour group,5-hour group and 6-hour group,with 5 rabbits in each group,and amiodarone was instilled into the other side of the margin ear vein every 1 hour,2 hours,3 hours,4 hours,5 hours and 6 hours.The degree of phlebitis of ear margin was observed.ELISA was used to detected the C-reactive protein(CRP) concentration.HE staining was used to determine the degree of inflammation.TUNEL was used to detect the apoptosis level of vascular endothelial cells.Results In group A,there was no visible change in the margin ear vein of rabbit in the 1-to 4-hour groups;the infusion site of 1 large-eared white rabbit in 5-hour group was red and swollen,while the blood vessel of 1large-eared white rabbit in 6-hour group was slightly stringlike.The was no statistically significant difference in C-reactive protein(CRP) concentration among the 1 h,2 h,and 3 h groups of group A(P>0.05);and the CRP concentrations gradually increased in the 4-hour,5-hour,and 6-hour groups(P<0.05).The results of HE staining and TUNEL staining showed that the vascular inflammatory injury was minimal within 4 hours in group A,while the inflammatory injury gradually aggravated from 4 hours onwards,and the endothelial cell apoptosis also significantly increased.In group B,injury inflammatory and cell apoptosis were improved after 72 hours of repair.In group C,the degree of vascular inflammatory injury and c
