摘  要:目的:研究外源性线粒体移植是否能减轻皮肤成纤维细胞受紫外线刺激引起的凋亡。方法:从骨髓间充质干细胞(BMMSCs)中提取线粒体,与皮肤成纤维细胞共培养使其移植进入细胞,JC-1染色明确线粒体浓度、治疗时间对细胞内最终线粒体数量、质量的影响。以1.5 J/cm 2与6 J/cm 2的紫外线照射分别模拟普通、极端两种刺激环境,流式细胞技术定量检测线粒体移植对皮肤成纤维细胞凋亡的影响。CCK-8验证外源性线粒体移植后对皮肤成纤维细胞存活的影响。结果:BMMSCs来源线粒体可移植进入皮肤成纤维细胞中,移植效率具有时间、浓度依赖性。并成功逆转紫外线刺激造成的线粒体膜电位下降,尤其在极端刺激环境中,可减轻细胞死亡、晚期与早期凋亡(P<0.001)。结论:外源性线粒体移植可显著减轻紫外线刺激引起的细胞凋亡,提升细胞的存活率。Objective:To investigate the potential of exogenous mitochondrial transplantation in reducing apoptosis of skin fibroblasts stimulated by ultraviolet light(UV).Methods:Mitochondria were extracted from bone marrow mesenchymal stem cells(BMMSCs)and co-cultured with skin fibroblasts.The concentration and treatment duration of mitochondria on the transplantation efficiency was assessed using JC-1 staining.Flow cytometry was employed to quantitatively measure the effects of mitochondrial transplantation on the apoptosis of skin fibroblasts simulated by UV at 1.5 J/cm 2 and 6 J/cm 2 respectively.CCK-8 assay was used to verify the effect of mitochondrial transplantation on the survival of the cells.Results:Successful transplantation of mitochondria derived from BMMSCs into skin fibroblasts effectively mitigated the decrease in mitochondrial membrane potential caused by UV stimulation in a time-and concentration-dependent manner.Particularly under UV of 6 J/cm 2,reduced cell death,late-stage apoptosis,early-stage apoptosis and restored the number of surviving cells(P<0.001).Conclusion:Exogenous mitochondrial transplantation can alleviate apoptosis caused by UV stimulation and increase the survival rate of cells.
