摘  要:目的分析子痫前期(PE)与正常妊娠孕妇血清中代谢物的差异,寻找潜在诊断代谢分子。方法选取2022年1月至12月徐州市妇幼保健院的PE患者6例和正常妊娠孕妇10例,应用超高液相色谱-串联质谱法检测入组孕妇血清样本,筛选差异性代谢物进行分析。结果共筛选出β-烟酰胺单核苷酸、D-(+)-甘露糖、溶血磷脂酰丝氨酸(Lysops)22:6、溶血磷脂酰胆碱(LPC)18:2、氧化型磷脂酰乙醇胺(OxPE)18:0+20:4+1O(1Cyc)、溶血磷脂酰乙醇胺(LPE)18:2、LPE 20:4、肌酸、十四烷酸、1-甲基尿酸、7-甲基黄嘌呤、磷脂酰胆碱(PC)16:0/18:2及鞘磷脂(SM)d14:0/20:0等13种差异性代谢物,ROC曲线分析结果显示十四烷酸、1-甲基尿酸、PC 16:0/18:2、7-甲基黄嘌呤及SM d14:0/20:0的曲线下面积大于0.7。结论PE患者与正常孕妇相比血清中代谢成分具有差异,代谢分子对于PE具有潜在诊断价值。Objective To assess the serum metabolite levels in preeclampsia(PE)and normal pregnant women,and search metabolites with potential diagnostic value.Methods Six cases of PE patients and 10 cases of normal pregnant women in Xuzhou Maternity and Child Health Care Hospital were selected from January to December 2022.Serum metabolites were detected by ultra-high performance liquid chromatography-mass spectrometry,followed by the analysis of the differential metabolites.Results A total of 13 differential metabolites were found,including betanicotinamide mononucleotide,D-(+)-mannose,lysophosphatidylserine(Lysops)22:6,lysophosphatidylcholine(LPC)18:2,oxidized phosphatidylethanolamine(OxPE)18:0+20:4+1O(1Cyc),lysophosphatidylethanolamine(LPE)18:2,LPE 20:4,creatine,tetradecanedioic acid,1-methyluric acid,7-methylxanthine,phosphatidylcholine(PC)16:0/18:2,and sphingomyelin(SM)d14:0/20:0.Receiver operating characteristic curve analysis showed that areas under the curve of tetradecanedioic acid,1-methyluric acid,7-methylxanthine,PC 16:0/18:2,and SM d14:0/20:0 were above 0.7.Conclusion There are differences in the serum metabolite levels between PE and normal pregnant women.The serum metabolites exhibit the diagnostic potential for PE.
