摘 要:目的观察六味地黄汤对多囊卵巢综合征(PCOS)大鼠性激素及转化生长因子β/Smads(TGF-β/Smads)信号通路相关因子的影响。方法随机将20只雌性SD大鼠分为空白对照组5只,造模组15只。造模组大鼠予来曲唑溶液8 mL/kg灌胃,连续21天造模,同时空白对照组予等量1%羧甲基纤维素溶液(CMC)灌胃,造模成功后将造模组大鼠进一步分为模型组、中药组、中西医结合组,每组5只。中药组大鼠予六味地黄汤生药7.5 g/(kg·d)灌胃,中西医结合组大鼠在中药组基础上予炔雌醇环丙孕酮溶液10 mL/(kg·d)灌胃,模型组和空白对照组予0.9%氯化钠注射液7.5 mL/(kg·d)灌胃,均连续21天。用酶联免疫吸附法检测各组大鼠血清黄体生成激素(LH)、卵泡刺激素(FSH)、雌二醇(E_(2))、睾酮(T)水平,蛋白免疫印迹法检测各组大鼠卵巢组织TGF-β_(1)、Smad3、Smad4、Smad7蛋白表达。结果与空白对照组比较,模型组LH、T水平升高(P<0.05);与模型组比较,中药组、中西医结合组LH、T水平均降低(P<0.05),而中药组LH、T水平与中西医结合组比较差异无统计学意义(P>0.05)。4组FSH、E_(2)比较差异均无统计学意义(P>0.05)。与空白对照组比较,模型组TGF-β_(1)、Smad3、Smad4、Smad7蛋白表达水平均升高(P<0.05);与模型组比较,中药组TGF-β_(1)、Smad3、Smad4蛋白表达水平均降低(P<0.05),Smad7蛋白表达水平升高(P<0.05),中西医结合组Smad3、Smad4、Smad7蛋白表达水平均降低(P<0.05),而TGF-β_(1)蛋白表达水平与模型组比较差异无统计学意义(P>0.05)。与中西医结合组比较,中药组TGF-β_(1)蛋白表达水平降低(P<0.05),Smad3、Smad4、Smad7蛋白表达水平升高(P<0.05)。结论六味地黄汤能改善PCOS大鼠的性激素水平,调节TGF-β/Smads信号通路相关因子的表达。Objective To observe the effect of Liuwei Dihuang Decoction on sex hormone in rats with polycystic ovarian syndrome(PCOS)and its influence on transforming growth factorβ/Smads(TGF-β/Smads)signaling pathway.Methods Twenty female Sprague-Dawley(SD)rats were randomly assigned into the control group(n=5)and the modeling group(n=15),in which they were gavaged letrozole solution(8 mL·kg^(-1))and an equal amount of 0.1%carboxymethyl cellulose(CMC)solution for 8 weeks,respectively.After successful modeling,the rats in the modeling group were equally divided into model group(7.5 mL·kg^(-1)·d^(-1)0.9%sodium chloride),traditional Chinese medicine(TCM)group(7.5 g·kg^(-1)·d^(-1) crude drugs of Liuwei Dihuang Decoction),and integrative medicine(IM)group(10 mL·kg^(-1)·d^(-1) ethinylestradiol-cyproterone acetate+7.5 g·kg^(-1)·d^(-1) crude drugs of Liuwei Dihuang Decoction).Treated for 21 days,the serum luteinizing hormone(LH),follicle-stimulating hormone(FSH),estradiol(E_(2)),testosterone(T)were detected with enzyme-linked immunosorbent assay(ELISA),the protein expressions of transforming growth factorβ_(1)(TGF-β_(1)),Smad3,Smad4 and Smad7 were detected by Western Blot.Results The levels of LH and T in the model group were significantly higher than those in the control group(P<0.05),which in the TCM group and IM group were significantly lower than the model group(P<0.05).The difference was not statistically significant in LH and T levels between the TCM group and the IM group(P>0.05).None of the FSH and E_(2) differed significantly among the four groups(P>0.05).Compared with the control group,the model group TGF-β_(1)、Smad3、Smad4、Smad7 were higher(all P<0.05).Compared with the model group,the TCM group presented significantly lower TGF-β_(1),Smad3,Smad4 and higher Smad7(all P<0.05),the IM group showed significantly lower Smad3,Smad4,Smad7(P<0.05).None of the TGF-β_(1) differed significantly between the IM group and the model group(P>0.05).The TCM group presented significantly lower TGF-β_(1) and higher Sma
