摘 要:采用蒽酮-硫酸比色法,在620 nm波长下,单因素分别考察显色剂浓度、显色剂用量、显色时间、显色温度,利用响应面法优化乌天麻粗多糖的测定条件.结果可知,影响吸光度因素顺序为显色剂用量>显色温度>显色时间.乌天麻粗多糖测定的最佳条件为显色剂质量浓度3.0 mg/mL、剂用量4.2 mL、温度91℃、时间10 min.预测值(0.5559)与实验值(0.5584)基本吻合.优化条件下显色测得昭通彝良乌天麻粗多糖含量最高(35.21%),其余5个产地粗多糖含量均低于25.00%.在本实验条件下,标准曲线呈良好的线性关系,精密度高、稳定且回收率好,是乌天麻粗多糖含量测定的最佳方法.The content of polysaccharide was determined by anthrone-sulfuric acid methods at 620 nm wavelength,the concentration,dosage,time and temperature of the chromogenic agent were investigated by single factor.The response surface method was used to establish and optimize the determination conditions of crude polysaccharide from Gastrodia elata f.glauca.The order of factors affecting the absorbance was as follows:the dosage of chromogenic agent>coloration temperature>coloration time.The optimal conditions for the determination of crude polysaccharide from Gastrodia elata f.glauca were as follows:concentration of chromogenic agent was 3.0 mg/mL,dosage of chromogenic agent was 4.2 mL,coloration temperature was 91℃,and coloration time was 10 min.The predicted value(0.5559)was basically consistent with the experimental value(0.5584).The content of crude polysaccharide from Zhaotong Yiliang Gastrodia elata f.glauca was the highest(35.21%),and the content of crude polysaccharide in the other five producing areas were lower than 25.00%.Under the conditions of this expriment,the standard curve showed a good linear relationship,high precision,stability and good recovery,which was the best method for the determination of crude polysaccharide content in Gastrodia elata f.glauca.Under the experimental conditions,the standard curve showed a good linear relationship,high precision,stability and good recovery.It was the best method for the determination of crude polysaccharide content of Gastrodia elata f.glauca.
