摘 要:烟叶原料的稳定性、一致性是保障烟草工业公司产品高质量发展的关键,有效区分和鉴别烟叶原料是当前工业公司亟需解决的难点。本研究以江苏中烟核心原料产区主栽的23个烤烟品种为材料,利用聚丙烯酰胺凝胶电泳技术,从212对SSR引物中最终筛选出12对多态性高且条带清晰的引物,等位基因数介于2~5,多态性信息含量介于0.3338~0.5711。聚类分析发现,各品种按照遗传背景聚为4类;群体遗传结构和主成分分析均将供试材料划分为2个亚群,且类群内各品种的分布结果高度一致,与种质亲缘关系吻合度较高。该组引物高效可靠,可用于烤烟品种的DNA指纹图谱构建和遗传多样性分析。本研究可为江苏中烟核心产区主栽烤烟品种的育种利用和烟叶原料鉴定提供理论依据。The stability and consistency of tobacco leaf raw materials are key for ensuring the high-quality development of tobacco industrial companies’products.Effective differentiation and identification of tobacco leaf raw materials is an urgent challenge that industrial companies need to solve.In this study,23 flue-cured tobacco varieties mainly planted in the production areas of Tobacco Jiangsu Industrial Corporation were used as materials.Eventually,12 pairs of primers with high polymorphism and clear bands were selected from 212 pairs of SSR primers by polyacrylamide gel electrophoresis detection.The number of alleles ranged from 2 to 5,and the polymorphism information content ranged from 0.3338 to 0.5711.Cluster analysis revealed that each variety was clustered into four categories based on genetic background.Based on the population genetic structure and principal component analysis the tested materials were divided into two subgroups.The distribution results of varieties within the groups were highly consistent,and were largely agreed with the pedigree relationship.This set of primers was highly efficient and reliable,and it could be used for constructing DNA fingerprints and genetic diversity analysis of flue-cured tobacco varieties.The results from this study could provide theoretical basis for the breeding utilization and identification of tobacco leaf materials of main flue-cured tobacco varieties in the Tobacco Jiangsu Industrial Corporation core production areas.
