摘 要:目的建立快速准确测定血液和尿液中总汞含量的电感耦合等离子体质谱法,为汞中毒的诊断提供技术支持。方法将血液和尿液用0.1%(V/V)Triton X-100-0.5g/LEDTA-2%(V/V)氨水混合溶液直接稀释50倍,电感耦合等离子体质谱仪测定其汞含量。选用铋为内标元素,校正实验过程中信号漂移带来的干扰和样品基质引起的基体效应。结果此方法的线性范围为0μg/L~3.00μg/L,检出限为0.51μg/L,回收率为91.8%~106.0%,样品测定的精密度<5%。采用本方法分析了汞中毒患者治疗期间血样和尿液中汞含量的变化。结论本方法快速简便,其准确度和精密度均符合检测的要求,适用于血液尿液中汞含量的日常监测、汞中毒的诊断和治疗过程中汞含量的检测。Objective This paper aims to establish a rapid and accurate method for the determination of total mercury in blood and urine by inductively coupled plasma mass spectrometry(ICP-MS/MS),so as to provide technical support for the diagnosis of mercury poisoning.Methods Blood and urine were directly diluted for 50 times with 0.1%(V/V)Triton X-100-0.5 g/L EDTA-2%(V/V)ammonia mixed solution,and then the mercury content was determined by ICP-MS/MS.Bismuth was selected as the internal standard element to correct the interference caused by instrument signal drift and the matrix effect caused by the sample matrix during the experiment.Results The linear range of this method was within Oμg/L-3.00μg/L.The detection limit was 0.51μg/L.The recovery rate was within 91.8%-106.0%.The precision was less than 5%.This method was used to analyze the changes of mercury content in blood and urine samples during the treatment of patients with mercury poisoning.Conclusion This method is fast and simple,and its accuracy and precision meet the requirements of detection,which is suitable for daily monitoring of mercury content in blood and urine,diagnosis of mercury poisoning and detection of mercury content in the treatment.
