摘 要:目的探讨参丹方对扩张型心肌病(DCM)模型大鼠心脏的保护作用及机制。方法用腹腔注射2.5 mg·kg^(-1)阿霉素建立DCM模型大鼠。将模型大鼠随机分为模型组和低、中、高剂量实验组,每组8只;另取8只正常大鼠作为正常组。低、中、高剂量实验组分别灌胃给予1.46、2.92、5.84 g·kg^(-1)参丹方溶液,剂量以10 mL·kg^(-1)·d^(-1)计算,正常组和模型组均灌胃给予等量0.9%NaCl。5组大鼠每日给药1次,连续给药28 d。用酶联免疫吸附试验法检测N末端脑钠肽前体(NT-proBNP)、肌钙蛋白T(cTn-T)和肌酸激酶(CK)的含量,用蛋白质印迹法检测磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)蛋白的表达情况。结果中、高剂量实验组和模型组、正常组的NT-proBNP水平分别为(963.40±46.67)、(903.11±82.76)、(1457.56±66.28)和(789.79±40.53)pg·mL^(-1),cTn-T水平分别为(559.68±31.05)、(521.51±19.29)、(685.53±17.16)和(452.67±30.76)pg·mL^(-1),CK水平分别为(39.23±6.85)、(34.61±8.55)、(55.79±0.61)和(27.79±1.76)ng·mL^(-1),PI3K蛋白相对表达水平分别为2.62±0.21、3.30±0.10、0.76±0.24和3.54±0.16,AKT蛋白相对表达水平分别为2.20±0.36、3.13±0.32、0.92±0.24和3.79±0.19。模型组的上述指标与中、高剂量实验组相比较,差异均有统计学意义(均P<0.01)。结论参丹方可能通过PI3K/AKT信号通路,改善DCM大鼠心肌细胞凋亡及心肌纤维化。Objective To study the protective effect of Shendan prescription on the heart of dilated cardiomyopathy(DCM)model rats and its mechanism.Methods DCM model rats were established by intraperitoneal injection of 2.5 mg·kg^(-1) adriamycin.The model rats were randomly divided into model group and experimental-L,-M,-H groups,with 8 rats in each group.Another 8 normal rats were selected as the normal group.Experimental-L,-M,-H groups were given 1.46,2.92 and 5.84 g·kg^(-1) Shendan prescription solution by intragastric administration,respectively,and the dose was calculated as 10 mL·kg^(-1)·d-1.Normal group and model group were given the same amount of 0.9%NaCl by intragastric administration.Five groups of rats were given the drug once a day for 28 days.The contents of N-terminal bra in natriuretic peptide precursor(NT-proBNP),cardiac troponin T(cTn-T)and creatine kinase(CK)were detected by enzyme-linked immunosorbent assay.The expression of phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)protein was detected by Western blot.Results The levels of NT-proBNP in the experimental-M,-H groups,model group and normal group were(963.40±46.67),(903.11±82.76),(1457.56±66.28)and(789.79±40.53)pg·mL^(-1);the levels of cTn-T were(559.68±31.05),(521.51±19.29),(685.53±17.16)and(452.67±30.76)pg·mL^(-1);the levels of CK were(39.23±6.85),(34.61±8.55),(55.79±0.61)and(27.79±1.76)ng·mL^(-1);the relative expression levels of PI3K protein were 2.62±0.21,3.30±0.10,0.76±0.24 and 3.54±0.16;the relative expression levels of AKT protein were 2.20±0.36,3.13±0.32,0.92±0.24 and 3.79±0.19,respectively.The above indexes in the model group were statistically significant compared with those in the experimental-M,-H groups(all P<0.01).Conclusion Shendan prescription may improve myocardial apoptosis and myocardial fibrosis in DCM rats through PI3K/AKT signaling pathway.
