摘 要:目的运用网络药理学研究方法对参芪地黄汤治疗糖尿病视网膜病变(diabetic retinopathy,DR)主要活性成分及核心基因进行分析。方法采用中药系统药理学分析平台筛选出参芪地黄汤组成药物的有效活性成分及作用靶点基因,通过Gene Cards数据库收集DR疾病基因,然后对药物作用基因及疾病相关基因进行韦恩分析,确定药物作用潜在基因,进而构建蛋白相互作用和活性成分-基因相互作用网络,并进行细胞功能注释和通路富集分析。结果研究得到了如槲皮素、山奈酚、豆甾醇、β-谷甾醇和异鼠李素等179种可直接作用于DR疾病基因的活性成分,以及丝氨酸/苏氨酸激酶1、肿瘤坏死因子-α、白细胞介素-1β、白细胞介素-6、血管内皮生长因子等143个潜在基因,并通过通路富集分析确定了101条作用通路,细胞功能注释确定了976种细胞功能。结论参芪地黄汤中的药物活性成分,可通过调节DR病程中的炎症反应对DR产生治疗效果。Objective To use network pharmacology research methods to analyze the main active ingredients and core genes of Shenqi Dihuang Decoction in treating diabetic retinopathy(DR).Methods The traditional Chinese medicine systems pharmacology analysis platform was used to screen out the effective active ingredients and target genes of the drugs composed of Shenqi Dihuang Decoction.DR disease genes were collected through the Gene Cards database,and then Wayne analysis was performed on drug-action genes and disease-related genes to determine potential genes for drug action.Then,protein interaction and active ingredient-gene interaction networks were constructed,and cell function annotation and pathway enrichment analysis were performed.Results The study obtained 179 active ingredients that can directly act on DR disease genes,such as quercetin,kaempferol,stigmasterol,β-sitosterol and isorhamnetin.As well as 143 potential genes such as serine/threonine kinase 1,tumor necrosis factor-α,interleukin-1β,interleukin-6,and vascular endothelial growth factor.And 101 action pathways were identified through pathway enrichment analysis,and 976 cell functions were identified through cell function annotation.Conclusion The active pharmaceutical ingredients in Shenqi Dihuang Decoction can produce therapeutic effects on DR by regulating the inflammatory response in the course of DR.
