摘  要:目的基于数据挖掘及网络药理学研究手段,总结国医大师郭诚杰治疗乳腺增生病(Mammary gland hyperplasia,MGH)的用药规律特点,并对核心药对进行药理机制分析。方法整理国医大师郭诚杰在陕西中医药大学附属医院2011年3月—2017年3月诊治的MGH医案,运用SPSS molder、SPSS Statistics系统对其临床处方用药规律进行研究,网络药理学分析最高支持度药组治疗MGH的成分、靶点、作用通路等。用Autodock系统进行分子对接。结果共纳入689首处方,处方中多使用补虚药、理气药、活血化瘀药,药性寒、温居多,苦、甘、辛药味为主,注重肝、脾两经的药物配伍使用;香附-当归是郭诚杰教授治疗MGH的核心药对;香附-当归对药的重要活性成分有槲皮素、山奈酚、木犀草素、豆甾醇、β-谷甾醇等;药对参与疾病共有靶点154个;主要信号通路有IL-18信号通路、AGE-RAGE信号通路等。分子对接验证显示,活性槲皮素与关键靶点成分TP53的对接得分最高,亲和力最好。结论郭诚杰教授治疗MGH主张以肝为本,在这一原理的指导下,以疏肝理气、活血化瘀为治则,核心药对香附-当归可能通过IL-18信号通路、AGE-RAGE信号通路起到治疗MGH的作用。Objective To summarize the characteristics of the medication pattern of Guo Chengjie,a national medical master,in the treatment of mammary gland hyperplasia(MGH)based on data mining and network pharmacology re⁃search to ols and to analyze the pharmacological mechanism of the core drug pairs.Methods The medical cases of MGH treated by Guo Chengjie,a national medical master,at the Affiliated Hospital of Shaanxi University of Traditional Chinese Medicine from March 2011 to March 2017 were collelted,The clinical prescription medication patterns were studied using SPSS molder and SPSS Statistics system.The components,targets,and pathways of action of the highest support drug group for the treatment of MGH were analyzed by network pharmacology.Molecular docking was performed using the Autodock system.Results A total of 689 prescriptions were included,which mostly used deficiency tonics,qi-regulating drugs,and blood-activating drugs,with cold and warm drug properties predominating and bitter,sweet,and pungent drug flavors pre⁃dominating,focusing on the use of drug combinations of liver and spleen meridians;Rhizoma Cyperi-Radix Angelicae Sinensis was the core drug pair used by Prof.Guo Chengjie for the treatment of this disease;the important active ingredients of Rhizoma Cyperi-Radix Angelicae Sinensis drug pair included quercetin,kaempferol,lignan,dousterol,β-sitosterol,etc.;there were 154 targets for the drug pair to regulate the diseases.The main signaling pathways included the IL-18 sig⁃naling pathway,AGE-RAGE signaling pathway,etc.Molecular docking validation showed that active quercetin had the highest docking score and the best affinity with the key target component TP53.Conclusion Professor Guo Chengjie advo⁃cates the treatment of MGH with the liver as the basis,and under the guidance of this principle,the core drug pair of Rhizo⁃ma Cyperi-Radix Angelicae Sinensis may play a role in the treatment of MGH through the IL-18 signaling pathway and AGE-RAGE signaling pathway by draining the liver,regulating
