基于数据挖掘及网络药理学补肾疏肝法治疗多囊卵巢综合征的用药规律及机制分析

摘　　要：目的 分析补肾疏肝法治疗多囊卵巢综合征(PCOS)的用药规律,挖掘核心中药治疗PCOS的药理学作用机制。方法 以中国知网、万方数据库、维普数据库、中国生物医学文献数据库作为文献来源,检索时间范围为2005年1月至2022年12月。采用数据挖掘技术对方药进行关联规则、系统聚类分析等。筛选核心中药的作用靶点及PCOS的治疗靶点;构建交集靶点蛋白-蛋白相互作用网络进行拓扑学分析筛选核心靶点;对交集靶点进行GO和KEGG通路富集分析,最终得出核心成分。结果 8味核心中药包括白芍、当归、菟丝子、熟地黄、香附、柴胡、郁金、山茱萸,其中柴胡-白芍、熟地黄-山茱萸为核心药对。网络药理学分析得出198个共同靶点,主要涉及胰岛素抵抗、类固醇激素生物合成相关的多个信号通路,包括丝氨酸/苏氨酸蛋白激酶1(AKT1)、丝裂原活化蛋白激酶3(MAPK3)、丝裂原活化蛋白激酶1(MAPK1)、雌激素受体1(ESR1)、AP-1转录因子(FOS)、白介素-6(IL-6)、肿瘤坏死因子(TNF)等关键靶点。结论 补肾疏肝法治疗PCOS用药以平补肝肾、柔肝疏肝为主,少辅健脾、活血之品。8味核心中药主要从改善胰岛素抵抗、抗炎、抗氧化等方面协同发挥抗PCOS的药理作用。Objective To explore the medication rules of Bushen Shugan method through data mining and analyze the pharmacological mechanism of core drugs in the treatment of polycystic ovary syndrome based on network pharmacology.Methods Using CNKI,Wanfang,VIP and China Biomedical Literature Database as literature sources,the search period was from January 2005 to December 2022.Data mining technology was used to conduct association rules and systematic cluster analysis for drugs.Screening the intersection target,constructing the intersection target protein-protein interaction network and screening the core targets by topological analysis.GO and KEGG pathway enrichment analysis was performed for intersection targets,and the core components were obtained.Results The 8 core medications were:Baishao,Danggui,Tusizi,Shudihuang,Xiangfu,Chaihu,Yujin,Shanzhuyu.Among them,Chaihu-Baishao,Shudihuang-Shanzhuyu are the core drug pairs.Network pharmacology analysis revealed 198 common targets,mainly related to insulin resistance,steroid hormone biosynthesis and other signaling pathways,including key targets such as AKT1,MAPK3,MAPK1,ESR1,FOS,IL-6,TNF,etc.Conclusions In the treatment of PCOS,the Bushen Shugan method mainly focuses on tonifying the liver and kidney,softening and soothing the liver,supplemented by tonifying the spleen and promoting blood circulation.The 8 core drugs mainly synergistically play the pharmacological effects of anti-PCOS in the aspects of improving insulin resistance,anti-inflammatory and antioxidant.
