摘 要:为了探究筛孔蜈蚣属、卫蜈蚣属和蜈蚣属的线粒体基因组序列特征,了解蜈蚣亚科和耳孔蜈蚣亚科在蜈蚣目中的系统发育关系,通过高通量测序技术、序列组装和基因组注释,新获得蜈蚣目蜈蚣科3个物种—卫蜈蚣属长足卫蜈蚣(Rhysida longipes(Newport,1845))、筛孔蜈蚣属筛孔蜈蚣sp.1(Ethmostigmus sp.1)和蜈蚣属日本蜈蚣(Scolopendra japonica L.Koch,1878)的全长线粒体基因组序列,其长度分别为14992、15002和14425 bp.通过线粒体基因组注释和分析发现,长足卫蜈蚣、筛孔蜈蚣sp.1和日本蜈蚣线粒体基因组13个蛋白质编码基因(PCG)的排列顺序均为COX1-COX2-ATP8-ATP6-COX3-NAD3-NAD5-NAD4-NAD4L-NAD6-COB-NAD1-rrnL-rrnS-NAD2,但筛孔蜈蚣sp.1线粒体基因组中NAD5、NAD4、NAD4L、NAD1、rrnL、rrnS的转录方向与其他序列相反.基于13个PCGs联合的14个唇足纲物种的系统发育树显示,长足卫蜈蚣与筛孔蜈蚣sp.1的关系紧密,与日本蜈蚣的关系则较远,地蜈蚣目(Geophilomorpha)和蜈蚣目(Scolopendromorpha)互为姊妹群.In order to explore the mitochondrial genome sequence characteristics of the genera Rhysida,Ethmostigmus and Scolopendra,and to clarify the phylogenetic relationship between Scolopendrinae and Otostigminae,the full-length mitochondri-al genome sequence of three species,i.e.Rhysida longipes(Newport,1845),Ethmostigmus sp.1 and Scolopendra japonica L.Koch,1878 was obtained using the methods of High-throughput sequencing techniques,assembly and genome annotation.The length of the three complete mitochondrial genome was 14992,15002 and 14425 bp,respectively.Through the annotation and analysis of the mitochondrial genome,the order of the 13 protein-coding genes was found to be COX1-COX2-ATP8-ATP6-COX3-NAD3-NAD5-NAD4-NAD4L-NAD6-COB-NAD1-rrnL-rrnS-NAD2 in R.longipes,Ethmostigmus sp.1 and S.japonica,but the direction of 6 protein-coding genes(NAD5,NAD4,NAD4L,NAD1,rrnL,rrnS)was oppsite in Ethmostigmus sp.1.Besides,the phylogenetic relationships of 14 species within Chilopoda were established based on 13 protein coding genes from mitochondrial genomes.The result showed that R.longipes had a close relationship with Ethmostigmus sp.1,and the relation-ship with S.japonica was faraway.Geophilomorpha and Scolopendromorpha were sister groups.
