摘 要:从天津滨海湿地土壤中分离筛选得到一株具有高效脱氮能力的异养硝化细菌WP5,通过16S rRNA序列分析,鉴定该菌株属于肠杆菌属(Enterbacter),将其命名为Enterbacter sp.WP5.以氨态氮为氮源,研究碳源、温度、氨氮负荷、接种量对该菌株生长和脱氮特性的影响,并应用正交实验探究其脱氮的优化条件.实验结果表明,碳源、温度、氨氮负荷、接种量对该菌株的生长和脱氮效能均有显著影响(P<0.05).正交实验结果显示,各因素对该菌株总氮去除效能的影响从大到小依次为:温度>碳源>氨氮负荷>接种量.最优脱氮条件为:碳源为蔗糖、温度35℃、氨氮负荷50 mg/L、接种量5%,在此条件下,该菌株达到最佳总氮去除率91.41%.菌株Enterbacter sp.WP5具有优良的异养硝化特性,在治理滨海湿地氨氮污染方面将有很好的应用前景.A strain of heterotrophic nitrifying bacteria WP5 with high-efficient nitrogen removal ability was isolated from the soil samples of Tianjin coastal wetland.Through 16S rRNA sequence analysis,the strain was identified to belong to Enterbac-ter,and was named as Enterbacter sp.WP5.The effects of carbon source,temperature,initial ammonia nitrogen concentra-tion and inoculum amount on the growth and nitrogen removal characteristics of the strain were studied,and the optimum con-ditions for nitrogen removal were investigated through orthogonal experiment.The results showed that carbon source,tempera-ture,initial ammonia nitrogen concentration and inoculum amount had significant effects on the growth and nitrogen removal efficiency of the strain(P<0.05).The results from the orthogonal experiment showed that the effects of the factors on the remo-val efficiency for total nitrogen of the strain ranged from the largest to the smallest as following:temperature>carbon source>initial ammonia nitrogen concentration>inoculum amount.The optimum nitrogen removal conditions were as following:the carbon source was sucrose,the temperature was 35℃,the initial ammonia nitrogen concentration was 50 mg/L,and the in-oculum amount was 5%,the optimal total nitrogen removal rate was 91.41%.Enterbacter sp.WP5 had excellent heterotrophic nitrifying ability,and would be a good candidate strain for high-efficient biological nitrogen removal in coastal wetlands.
