摘  要:为找到快速测定珍稀中藏药材桃儿七种子生活力的测定方法,以野生植株种子为研究材料,采用2,3,5-氯化三苯基四氮唑染色法(TTC法),运用4因素4水平的正交试验L_(16)(4^(4))设计,研究不同浸种时间、染色温度、TTC溶液浓度和染色时间对桃儿七种子生活力的影响。结果表明,桃儿七种子生活力测定的最佳条件为30℃下浸种24 h,35℃于1%TTC溶液中染色3 h,种子胚部染成红色,生活力达到了95.6%。该测定方法操作简单、准确、高效,在合适的条件下反应速度快,可得到明显的试验结果。In order to find a method for rapid determination of seed vigor of Tibetan medicine Sinopodo⁃phyllum hexandrum,the seeds of wild plants were used as research materials,and 2,3,5-triphenyl tetrazolium chloride staining method(TTC method)was used.The orthogonal test L_(16)(4^(4))design of 4 factors and 4 levels was used to study the effects of different staining time,dyeing temperature,TTC solution concentration and staining time on the seed vigor of S.hexandrum.The results showed that the optimum conditions for the determination of seed vigor of S.hexandrum were dyeing at 30℃ for 24 h and staining at 35℃ in 1%TTC solution for 3 h.The embryo of the seed was dyed red,and the vigor reached 95.6%.The method is simple,accurate and efficient.Under appropriate conditions,the reaction speed is fast and obvious test results can be obtained.
