摘 要:G0/G1期开关基因2(G0/G1 switch 2,G0S2)在脂肪稳态、细胞增殖和肿瘤发生等生物学过程中扮演着重要角色,而有关G0S2基因在家禽抗病毒免疫中的研究相对甚少。为探讨G0S2基因在鸡巨噬细胞中的抗病毒作用及其机制,构建G0S2真核表达载体并转染鸡巨噬细胞系HD11细胞,通过荧光定量PCR和Western blot检测G0S2过表达对J亚群禽白血病病毒(ALV-J)复制水平、天然免疫基因TLR3和糖酵解关键调控基因MYC表达的影响。结果表明:G0S2过表达可显著抑制ALV-J在HD11细胞中增殖。G0S2显著上调HD11细胞内双链RNA(dsRNA)识别受体基因TLR3表达,并抑制糖酵解调控基因MYC表达,说明激活TLR3表达和抑制MYC表达可能是G0S2基因抑制ALV-J在巨噬细胞中增殖的重要机制。综上,该研究揭示了鸡G0S2在巨噬细胞中的抗病毒功能及其作用机制,为今后研究G0S2的功能及其宿主遗传抗性提供了参考依据。G0/G1 switch gene 2(G0S2) plays an important role in biological processes such as fat homeostasis, cell proliferation and tumorigenesis. However, there are relatively few studies on G0S2 gene in poultry antiviral immunity. In order to explore the antiviral effect and mechanism of G0S2 gene in chicken macrophages, this study constructed a G0S2 eukaryotic expression vector and transfected it into chicken macrophage cell line HD11 cells. The influence of G0S2 overexpression on the expression of avian leukosis virus subgroup J(ALV-J) replication, innate immunity gene TLR3 and the key glycolysis regulatory gene MYC were detected by fluorescence quantitative PCR and Western blot. The results showed that G0S2 overexpression could significantly inhibit the proliferation of ALV-J in HD11 cells. Mechanistically, G0S2 significantly up-regulated the expression of the dsRNA recognition receptor TLR3 and inhibited the expression of the glycolysis-regulated gene MYC in HD11 cells. This indicated that activation of TLR3 expression and inhibition of MYC may be the important mechanism for G0S2 to inhibit the proliferation of ALV-J in macrophages. This study revealed the antiviral function and mechanism of chicken G0S2 in macrophages, and provided a reference for future research on the function of G0S2 and its host genetic resistance.
