摘  要:为探讨栝楼瞿麦汤对治疗糖尿病肾病肾纤维化的作用机制,测定糖尿病肾病大鼠血糖、体重等生化指标,利用HE、PAS、Masson染色和免疫组化技术来评估肾纤维化情况。采用Cell Count Kit-8检测栝楼瞿麦汤对异常状态的HBZY-1细胞增殖的影响,同时检测TGF-β1/Smads信号通路中与miR-21-3p相关的miRNA表达及相关蛋白质和mRNA水平。结果表明:栝楼瞿麦汤可降低糖尿病肾病大鼠肾脏与体重比例的大小、血尿素(BUN)、血清肌酐(Scr)和24 h尿蛋白。栝楼瞿麦汤可明显降低TGF-β1、p-Smad3蛋白表达,上调Smad7蛋白表达;并降低HBZY-1细胞miR-21-3p miRNA、Smad3、TGF-β1和Collagen I mRNA的表达,上调Smad7 mRNA的表达。综上,栝楼瞿麦汤可通过miR-21-3p调节TGF-β1/Smads信号通路,能改善肾功能,延缓肾纤维化进展。In order to investigate the mechanism of Gua Lou Qu Mai Decoction(GLQMD) in the treatment of diabetic nephropathy with renal fibrosis, biochemical indices such as blood glucose and body weight were determined in diabetic nephropathy rats, and the status of renal fibrosis was evaluated by HE, PAS, Masson staining and immunohistochemical techniques. The effect of GLQMD on the proliferation of abnormal HBZY-1 cells was detected by Cell Count Kit-8, and the expression of miRNA associated with miR-21-3p and the levels of related proteins and mRNA in the TGF-β1/Smads signaling pathway were detected. The results showed that GLQMD reduced the kidney to body weight ratio size, blood urea(BUN), serum creatinine(Scr), and 24-hour urinary protein in diabetic nephropathy rats. In addition, TGF-β1 and p-Smad3 protein expressions were significantly decreased and Smad7 protein expression was up-regulated. GLQMD decreased the mRNA expressions of miR-21-3p miRNA, Smad3, Collagen I and TGF-β1 in HBZY-1 cells, and up-regulated the mRNA expression of Smad7. In conclusion, GLQMD can regulate TGF-β1/Smads signaling pathway through miR-21-3p, and can also improve renal function and delay the progression of renal fibrosis, which may provide a guidance in the treatment of diabetic nephropathy by traditional Chinese medicine.
