摘 要:目的探讨表儿茶素(EC)通过核因子E2相关因子2/抗氧化反应元件(Nrf2/ARE)通路对脊髓损伤(spinal cord injury,SCI)小鼠的抗炎和抗氧化作用。方法采用改良Allen’s击打法制备小鼠脊髓损伤模型,采用随机数字表法将小鼠分为对照组(Sham组)、脊髓损伤组(SCI组)、表儿茶素处理组(EC组),每组6只。通过称重受损脊髓含水量评价受损脊髓的水肿情况;通过小鼠Basso Mouse Scale(BMS)评分,评价损伤小鼠的运动功能;通过悬尾实验和糖水偏嗜试验评价小鼠的抑郁及焦虑状态;使用免疫荧光染色观察神经元的数量;使用酶联免疫吸附试验(ELISA)法检测白细胞介素(IL)-1β、IL-10的表达;采用蛋白质印迹法(Western blot)检测Nrf2蛋白和Kelch样ECH关联蛋白1(Keap1)的表达。组间比较采用t检验和单因素方差分析。结果SCI 3 d,EC组的脊髓含水量明显低于SCI组,差异有统计学意义[(73.91±0.95)%比(77.78±0.95)%,F=8.611,P<0.05];BMS评分表明,从第14天开始,EC组小鼠的运动能力改善优于SCI组,差异有统计学意义;SCI 28 d,EC组小鼠糖水偏嗜度高于SCI组,差异有统计学意义[(64.25±2.50)%比(51.98±3.27)%,t=-7.316,P<0.05]。SCI 28 d,EC组小鼠不动时间明显低于SCI组,差异有统计学意义[(202.83±6.97)s比(260.50±10.62)s,t=-11.123,P<0.05];SCI 7 d,EC组胶质纤维酸性蛋白(GFAP)阳性细胞明显低于SCI组,差异有统计学意义(159.00±9.12比209.50±14.15,t=7.347,P<0.05),EC组NeuN阳性细胞数明显高于SCI组,差异有统计学意义(108.17±12.48比54.83±15.55,t=-6.552,P<0.05);SCI 7 d,EC组抑炎因子IL-10的表达量明显高于SCI组,差异有统计学意义[(596.48±13.30)pg/ml比(483.55±16.14)pg/ml,t=13.224,P<0.05]。同时,EC组促炎因子IL-1β的表达量明显低于SCI组,差异有统计学意义[(44.02±4.28)pg/ml比(87.00±5.75)pg/ml,t=14.686,P<0.05];SCI 7 d,EC组小鼠脊髓组织中的Nrf2蛋白表达量高于SCI组,差异有统计学意义(1.08±0.14比0.78±0.10,t=-4.245,P<0.05),EC组Objective To investigate the anti-inflammatory and antioxidant effects of epicatechin(EC)on mice with spinal cord injury(SCI)through nuclear factor erythroid-2 related factor 2/antioxidant response element(Nrf2/ARE)pathway.Methods The modified Allen’s impact method was used to prepare the rat model of SCI,and the mice were divided into control group(Sham group),SCI group and EC-treated group(EC group)by random number table method(n=6).The edema of the injured spinal cord was evaluated by weighing the water content of the injured spinal cord.The motor function of injured mice was evaluated by Basso Mouse Scale(BMS)score.Depression and anxiety were evaluated by tail suspension test and sugar water preference test.The number of neurons was observed by immunofluorescence staining.The expressions of interleukin(IL-1β)and interleukin(IL-10)were detected by enzyme-linked immunosorbent assay(ELISA).Western blotting was used to detect the expression of Nrf2 protein and kelch-like ECH-associated protein 1.T-test and one-way analysis of variance were used for comparison between groups.Results At post-injury day 3,the spinal water content in EC group was significantly lower than that in SCI group,the difference was statistically significant[(73.91±0.95)%vs.(77.78±0.95)%,F=8.611,P<0.05].The BMS score showed that from the 14th day,the improvement of motor ability of mice in EC group was better than that in SCI group,the difference was statistically significant.At post-injury day 28,the sugar water preference in EC group was higher than that in SCI group,and the difference was statistically significant[(64.25±2.50)%vs.(51.98±3.27)%,t=-7.316,P<0.05].At post-injury day 28,the immobility time of mice in EC group was significantly lower than that in SCI group,and the difference was statistically significant[(202.83±6.97)s vs.(260.50±10.62)s,t=-11.123,P<0.05].At post-injury day 7,the number of glial fibrillary acidic protein(GFAP)positive cells in EC group was significantly lower than that in SCI group,the difference was st
