摘 要:试验采用单因素和正交试验设计,对裂褶菌胞外多糖培养基进行优化,并研究裂褶菌胞外多糖的生物活性。结果显示,高产胞外多糖培养基的配方为乳糖40 g/L、大豆蛋白胨8 g/L、3-吲哚丁酸1 mg/L、KH_(2)PO_42 g/L、MgSO_(4)·7H_(2)O 1 g/L,pH值6.0。优化后裂褶菌胞外多糖产量达到(16.01±0.38)g/L,为基础培养基的3.03倍。裂褶菌胞外多糖对羟基自由基和DPPH自由基的半数抑制浓度(IC_(50))分别为3.146、8.291 g/L;对大肠杆菌和金黄色葡萄球菌的IC_(50)分别为3.516、9.732 g/L;在相对湿度(RH)43%、60%、81%环境中,裂褶菌胞外多糖60 h的吸湿率分别为28.0%、42.0%、50.9%,48 h内的保湿率为82.5%。研究表明,优化后的培养基配方可明显提高裂褶菌胞外多糖产量,且裂褶菌胞外多糖有良好的抗氧化性、抑菌性、吸湿性及保湿性。The study used a single factor and orthogonal experiment design,to optimize the culture medium and study the biological activity of Schizophyllum commune exopolysaccharides.The results showed that the formula of the high yield exopolysaccharide medium was lactose 40 g/L,soybean peptone 8 g/L,3-indolebutyric acid 1 mg/L,KH_(2)PO_(4)2 g/L,MgSO_(4)·7H_(2)O 1 g/L,pH value 6.0.After optimization,the polysaccharide yield reached(16.01±0.38)g/L,which was 3.03 times higher that of base medium.The IC_(50)value of exopolysaccharides for hydroxyl radical and DPPH free radical were 3.146 and 8.291 g/L,respectively.The IC_(50)value for Escherichia coli and Staphylococcus aureus were 3.516 and 9.732 g/L,respectively.At the RH of 43%,60%and 81%,the moisture absorbance rate of Schizophyllum commune exopolysaccharide within 60 h was 28.0%,42.0%and 50.9%,respectively,and the moisture retention rate within 48 h was 82.5%.The study indicates that the optimized medium formula can significantly increase the polysaccharide yield,and the Schizophyllum commune exopolysaccharide has a good antioxidant,antibacterial,hygroscopicity and moisture retention.
