摘 要:苹果的柱型结构具有生长紧凑、节间短的特点,这种树形非常适合进行高密度种植,对果园的生产及管理具有重要意义。为了进一步研究GA_(3)和苹果柱型性状之间的关系,利用外源GA_(3)对普通型苹果和柱型苹果的新梢、野生型烟草和转MdCoL基因烟草株系进行处理,通过qRT-PCR分析MdCoL基因表达量和GA生物合成及信号转导相关基因表达量的变化。结果表明:与普通型苹果相比,柱型苹果中GA合成相关基因MdGA2oxI相对表达量降低,而MdGA20oxI基因和DELLA蛋白相关基因MdRGL2表达量升高;外源GA_(3)处理柱型苹果后,与对照相比,MdCoL和MdGA2oxI基因下调表达,MdGA20oxI和MdRGL2基因上调表达。外源GA_(3)处理转MdCoL基因烟草后,发现烟草株高增加、节间变长、叶绿素含量降低。综上所述:GA_(3)是影响苹果柱型性状形成的一个重要原因。The columnar structure of apple has the characteristics of compact growth and short internode.This kind of tree is very suitable for high-density planting,which is of great significance for the production and management of orchards.In order to further study the relationship between GA_(3) and columnar apple traits,exogenous GA_(3) was used to treat spring shoots of ordinary and columnar apples,wild type tobacco(WT)and MdCoL gene transfer tobacco lines,and the changes of MdCoL gene expression and GA biosynthesis and signal transduction related genes were analyzed by qRT-PCR.The results showed that compared with ordinary apples,the expression level of GA synthetic gene MdGA 2 oxI was lower in columnar apples,while the expression level of MdGA 20 oxI gene and DELLA-related gene MdRGL2 was higher.Compared with the control group,the expression of MdCoL and MdGA 2 oxI was down-regulated and the expression of MdGA 20 oxI and MdRGL2 was up-regulated after exogenous GA_(3) treatment.After exposed to exogenous GA_(3),compared to WT,the plant height and internode length of MdCoL transgenic tobacco increased,while chlorophyll content decreased.In conclusion,GA_(3) is an important factor affecting the formation of apple columnar traits.
