摘  要:随着同步辐射、自由电子激光等先进光源的快速发展,基于先进光源的X射线成像技术为单细胞研究带来了新的机遇.X射线成像具有纳米分辨、成像深度大、定量成像、多参数分析等优势,研究对象可以从完整的单个细胞扩展到特定的生物分子,实现从多个尺度获取单细胞多方面的结构以及功能相关生物学信息.本文综述了基于先进光源的X射线成像技术及其在多个单细胞成像领域的应用进展,包括不同类型细胞的形貌与结构成像、跨尺度的细胞结构成像及单细胞-外源物质相互作用等,覆盖了从完整细胞到局部结构乃至特定分子多个层次的单细胞成像研究实例.本文对各种技术对于单细胞成像分析的适用性及未来发展进行了讨论,为基于先进光源的X射线单细胞成像研究提供参考与启示.With the rapid development of advanced light sources,such as synchrotron radiation and X-ray free electron laser,X-ray imaging techniques have brought new opportunities for single-cell research.X-ray imaging provides unique advantages,including nanometer-resolution,large depth of field,quantitative imaging and multi-parameter analysis,and the imaging objects range from a complete single cell to specific biological molecules,enabling multi-scale acquisition of structural and functional information of a single cell.This review shows X-ray imaging techniques based on advanced light sources and their application progress in multiple single-cell imaging fields,including morphological and structural imaging of different types of cells,imaging of cross-scale cellular structures,and interactions between a single cell and exogenous substances.It covers single-cell imaging research examples at multiple levels,from complete cells to subcellular structures and finally to specific molecules.Additionally,this review discusses the applicability and future development of X-ray imaging techniques for single-cell imaging analysis,and provides references and inspirations for X-ray single-cell imaging research based on advanced light sources.
