摘  要:为了探究促性腺激素释放激素(GnRH)疫苗对公犬生殖功能的影响,试验将GnRH基因克隆至原核表达载体中,转化到大肠杆菌BL21(DE3)感受态细胞中,用终浓度为1 mmol/L IPTG诱导蛋白表达,通过SDS-PAGE和Western-blot对表达产物进行蛋白鉴定,将鉴定正确的GnRH蛋白纯化后制备成抗原。选取6只2~4月龄健康公犬随机分成2组,每组3只,其中免疫组每只犬免疫GnRH蛋白1 mg,共免疫3次,每次间隔4周,分别于免疫前和一免、二免、三免后14天采血,通过ELISA检测血清GnRH抗体及睾酮水平,并于最后一次采血结束后制备睾丸组织切片,观察睾丸组织形态。结果表明:3次免疫后免疫组GnRH抗体水平均显著高于对照组(P<0.05),睾酮水平均显著低于对照组(P<0.05),免疫组睾丸相对较小,曲精小管中生精细胞数量较少,管径较细。说明主动免疫GnRH疫苗抑制了公犬的性腺发育和生殖器官的成熟。In order to study the effects of Echinops latifolius flavonoids on blood physiological and biochemical indexes,organ indexes in Kunming mice,and to evaluate the safety of Echinops latifolius flavonoids,in this experiment,the 80 Kunming mice were divided into 4 groups with 20 mice in each group,namely the high-dose group(by gavage with Echinops latifolius flavonoids 400 mg/kg according to body weight),medium-dose group(by gavage with Echinops latifolius 200 mg/kg according to body weight),low-dose group(by gavage with Echinops latifolius 100 mg/kg according to body weight)and control group(0.5 mL/d of purified water);continuous medication lasted for 30 days.At the end of the experiment,mice were subjected to blood collection to determine blood physiological and biochemical indexes.Each group of mice was sacrificed;the organ indexes of mice were calculated,and pathological histological sections were prepared from liver and kidney,and the histological changes of the cases were observed.The results showed that there was no significant difference in blood routine indexes in the high,medium and low dose groups(P>0.05),and the average number of red blood cells in the high dose group was significantly higher than that in the control group(P<0.05).The hemoglobin contents in the high-dose group and the medium-dose group were significantly higher than that in the control group(P<0.05).The blood routine indexes of all groups were within the normal range.There were no significant differences in blood biochemical indexes between high,medium and low dose groups(P>0.05),and the total cholesterol content in the high dose group was significantly lower than that in the control group(P<0.05).The activities of aspartate aminotransferase and the contents of triacylglycerol in the high-dose group and the medium-dose group were significantly lower than those in the control group(P<0.05).There was no significant difference in organ indexes between the high,medium and low dose groups(P>0.05),but the liver,lung and heart indexes of mice in
