短期施有机肥对‘川草2号’老芒麦AMF种类及分布的影响

摘  要:为探究高寒地区短期施有机肥对禾本科牧草丛枝菌根真菌(Arbuscular mycorrhizal fungi,AMF)群落多样性及群落结构的影响,本研究以‘川草2号’老芒麦为试验材料,进行田间小区试验,设置施有机肥(F)与不施有机肥(CK)处理,在生长结束后采集根周和根际土壤用于分析微生物的组成,测序后利用FUNGuild筛选AMF的操作分类单元(Operational taxonomic units,OTU),对根周和根际土壤AMF群落多样性和组成,以及它们与环境因子的相关性进行分析。结果表明:‘川草2号’老芒麦共生的AMF群落共有7个属,根周土壤的物种数大于根际土壤;AMF群落多样性受土壤pH值和牧草鲜重影响最大,为极显著负相关关系(P<0.01);有机肥增加了根周土壤AMF的多样性和物种数,降低了根际土壤AMF的物种数,且降低了根周和根际土壤AMF优势菌属近明球囊霉属(Claroideoglomus)的相对丰度。该结果为高寒地区牧草种植地田间养分管理和AMF多样性之间关系提供了理论依据。In order to explore the effect of short-term application of organic fertilizer on the diversity and community structure of arbuscular mycorrhizal fungi(AMF)associated with gramineous forage in alpine region,this study used Elymus sibiricus L.‘Chuancao No.2’as the experimental material,conducted a field plot experiment,and set up the treatments of organic fertilizer application(F)and no organic fertilizer(CK).Around the root soil and rhizospheric soil samples were collected at the end of growth period to analyze microbial composition.After sequencing,FUNGuild was used to screen the operational classification units of AMF,and then the diversity and composition of AMF community around the root soil and rhizosphere soil,as well as its correlation with environmental factors were analyzed.The results showed that there were 7 genera in the AMF community of Elymus sibiricus L.‘Chuancao No.2’,and the number of species around the root soil was exceed that in rhizospheric soil.The diversity of AMF community was mostly affected by soil pH and forage fresh weight,which was significantly negatively correlated(P<0.01).Application of organic fertilizer increased the diversity and species number of AMF around the root soil,decreased the species number of AMF in rhizospheric soil,and decreased the relative abundance of AMF dominant genus Claroideoglomus around both the root soil and in rhizospheric soil.The results could provide a theoretical basis for the relationship between field nutrient management and AMF diversity in alpine grassland.
