摘 要:目的 研究维生素D对紫外线诱导豚鼠皮肤晒伤的治疗作用,初步探索维生素D对紧密连接蛋白Claudins家族的作用机制。方法 雌性白化豚鼠36只,随机分成模型组、空白对照组、维生素D低剂量组、维生素D中剂量组、维生素D高剂量组、维生素D预防治疗组,每组6只。除空白对照组外,其余5组采用HGSS-Z型紫外太阳模拟器为辐射装置照射豚鼠脱毛区制备豚鼠晒伤模型。建模成功后,维生素D低剂量组(1500 IU/kg)、维生素D中剂量组(3000 IU/kg)、维生素D高剂量组(6000 IU/kg)、维生素D预防治疗组(6000 IU/kg,造模前7 d开始灌胃给药, 1次/d)均给予相应剂量维生素D灌胃,模型组和空白对照组给予等量生理盐水。给药7 d后,测量并计算创面愈合率,进行组织病理学检查并运用Western blot检测紧密连接蛋白Claudins家族的表达。结果 模型组、维生素D低剂量组、维生素D中剂量组、维生素D高剂量组、维生素D预防治疗组豚鼠皮肤在经过紫外线的照射后都出现了面积大小比较相近的红斑。给药7 d后,维生素D高剂量组红肿扩散得到控制,有散在的少量水泡;维生素D预防治疗组与模型组相比愈合情况明显更好,且出现结痂,结痂面积缩小。维生素D中剂量组豚鼠创面愈合率(36.62±9.18)%高于模型组的(20.57±8.12)%,差异具有统计学意义(P<0.05);维生素D高剂量组、维生素D预防治疗组豚鼠创面愈合率分别为(48.06±5.06)%、(63.76±7.13)%,显著高于模型组,差异具有统计学意义(P<0.01)。维生素D低剂量组豚鼠创面愈合率与模型组比较,差异无统计学意义(P>0.05)。模型组与空白对照组相比,豚鼠皮肤Claudin表达显著减少;维生素D低剂量组与模型组比较,豚鼠皮肤Claudin表达有所提高,但不明显;维生素D中剂量组、维生素D高剂量组、维生素D预防治疗组与模型组比较,豚鼠皮肤Claudin表达有显著提高,与组织学结果相符合。结论 维生素D对Objective To study the therapeutic effect of vitamin D on skin sunburn induced by ultraviolet rays in guinea pigs,and explore the mechanism of vitamin D on tight junction protein Claudins family.Methods A total of 36 female albino guinea pigs were randomly divided into model group,blank control group,low-dose vitamin D group,medium-dose vitamin D group,high-dose vitamin D group and vitamin D preventive treatment group,with 6 guinea pigs in each group.In addition to the blank control group,the sunburn model of guinea pigs of other 5 groups were irradiated with HGSS-Z UV solar simulator to prepare the guinea pig sunburn model.After successful modeling,low-dose vitamin D group(1500 IU/kg),medium-dose vitamin D group(3000 IU/kg),high-dose vitamin D group(6000 IU/kg),vitamin D preventive treatment group(6000 IU/kg,starting gavage administration 7 d before modeling,once a day)were given the corresponding dose of vitamin D by gavage,model group and blank control group were given the same amount of normal saline.After 7 d of administration,wound healing rate was measured and calculated,histopathological examination was performed,and the expression of Claudins family was detected by Western blot.Results The guinea pigs in the model group,the low-dose vitamin D group,the medium-dose vitamin D group,the high-dose vitamin D group and the prophylactic group all showed similar erythema after ultraviolet irradiation.After 7 d of administration,in the high-dose vitamin D group,the redness and swelling were controlled,and there were a few scattered blisters.Compared with the model group,the healing of vitamin D preventive treatment group was significantly better,and scab appeared and the area of scab was reduced.The wound healing rate of(36.62±9.18)%in medium-dose vitamin D group was higher than that of(20.57±8.12)%in model group,and the difference was statistically significant(P<0.05).The wound healing rates of high-dose vitamin D group and vitamin D preventive treatment group were(48.06±5.06)%and(63.76±7.13)%,which were sig
