摘 要:目的:研究青蒿琥酯(ART)对体外高糖环境下大鼠下颌下腺上皮细胞凋亡的影响。方法:通过体外培养、纯化并鉴定大鼠下颌下腺上皮细胞。使用CCK-8和western blotting检测不同浓度葡萄糖对大鼠下颌下腺上皮细胞增殖和功能的影响。使用含50 mmol/L葡萄糖的培养基模拟体外高糖环境,将细胞分为正常对照组(Con组)、高糖模型组(HG组)和不同浓度ART干预组(HG+5μmol/L ART组、HG+10μmol/L ART组、HG+20μmol/L ART组、HG+40μmol/L ART组)。分别采用实时荧光定量PCR(RT-qPCR)法和western blotting法检测细胞水通道蛋白5(AQP5)、Bax、Bcl-2和Caspase-3基因和蛋白表达。结果:随着高糖环境中葡萄糖浓度的增加,大鼠下颌下腺上皮细胞抑制率升高,AQP5蛋白表达水平降低(P<0.05)。与Con组相比,HG组细胞AQP5蛋白、Bcl-2基因和蛋白表达水平明显降低,Bax、Caspase-3基因和蛋白表达水平明显升高(P<0.05);与HG组相比,不同浓度ART干预组Bcl-2基因和蛋白表达水平及AQP5蛋白表达升高,Bax、Caspase-3基因和蛋白表达水平降低(P<0.05)。结论:ART对高糖环境下大鼠下颌下腺上皮细胞具有保护作用,其机制可能与抑制细胞凋亡有关。Objective:To investigate the effect of artesunate(ART)on apoptosis of rat submandibular gland epithelial cells in high glucose environment in vitro.Methods:Rat submandibular gland epithelial cells were cultured,purified and identified in vitro.The CCK-8 assay and western blotting were used to detect the effects of different concentrations of glucose on the proliferation and function of rat submandibular gland epithelial cells.The medium containing 50 mmol/L glucose was used to simulate the high glucose environment in vitro.The cells were divided into normal control group(Con group),high glucose model group(HG group)and different concentrations of ART intervention groups(HG+5μmol/L ART group,HG+10μmol/L ART group,HG+20μmol/L ART group,HG+40μmol/L ART group).The expression levels of aquaporin 5(AQP5),Bcl-2 associated X protein(Bax),B cell lymphoma/leukemia 2 protein(Bcl-2)and Caspase-3 were detected by real-time fluorescence quantitative PCR(RT-qPCR)and western blotting.Results:With the increase of glucose concentration in high glucose environment,the inhibition rate of rat submandibular gland epithelial cells increased and the protein expression level of AQP5 decreased(P<0.05).Compared with the Con group,the expression levels of AQP5 protein,Bcl-2 gene and protein in the HG group significantly decreased,while the expression levels of Bax,Caspase-3 gene and protein significantly increased(P<0.05).Compared with the HG group,the expression levels of Bcl-2 gene and protein and AQP5 protein in different concentrations of ART intervention groups increased,while the expression levels of Bax,and Caspase-3 gene and protein decreased(P<0.05).Conclusion:ART has a protective effect on rat submandibular gland epithelial cells in high glucose environment,and its mechanism may be related to the inhibition of apoptosis.
