摘 要:目的:以侧脑室注射法构建幼龄小鼠细菌性脑膜炎模型。方法:选取SPF级C57BL/6雄性小鼠5只,依据成年小鼠ICV坐标及发育期小鼠脑成像图谱,采用右侧脑室注射坐标,以0.5μL·min^(-1)的注射速度缓慢注射伊文思蓝染液2μL,15min后处死小鼠,剪开颅骨观察染液的扩散分布情况;另取SPF级C57BL/6雄性小鼠12只,随机分为对照组和模型组(n=6),以定位坐标(ML:-1.3mm,AP:-0.5mm,DV:-1.6mm)分别右侧脑室注射2μL含3.5×10^(5)CFU肺炎链球菌悬液(模型组)或生理盐水(对照组),20h后进行Loeffler神经行为学评分,并留取脑组织进行病理学评价。结果:侧脑室注射(坐标为ML:-1.3mm,AP:-0.5mm,DV:-1.6mm)法可成功构建4周龄小鼠肺炎链球菌脑膜炎模型,小鼠出现明显的神经行为学改变及脑膜炎性浸润和结构损伤。结论:参考本文方法可成功建立幼龄小鼠细菌性脑膜炎模型,用于探究儿童细菌性脑膜炎机制。Objective:To establish an immature mouse model of bacterial meningitis by intracerebroventricular injection.Methods:Five SPF male C57BL/6 mice were selected.Based on the coordinates for ICV in adult mice and the brain imaging atlas for developing mice,the injection coordinates for the right lateral ventricle were determined.A slow injection of 2μL of Evans Blue dye at 0.5μL·min^(-1) was performed.After 15 minutes,the mice were euthanized,and the distribution of the dye was observed after dissecting the skull.Another twelve SPF male C57BL/6 mice were randomly divided into control and model groups(n=6).In the model group,2μL of a suspension containing 3.5×10^(5) CFU of Streptococcus pneumoniae was injected into the right lateral ventricle at the following coordinates:ML:-1.3 mm,AP:-0.5 mm,DV:-1.6 mm.In the control group.In the control group,2μL of physiological saline was injected similarly.After 20 hours,the Loeffler neurological scoring test was conducted,and brain tissue was collected for pathological evaluation.Results:We successfully established a model of pneumococcal meningitis in 4-week-old mice using ICV(with coordinates:ML:-1.3 mm,AP:-0.5 mm,DV:-1.6 mm).The mice exhibited significant neurological behavior changes,meningeal infiltration,and structural damage.Conclusion:Referring to the methods described in this study,a successful immature mouse model of bacterial meningitis was established,which can be applied to investigate the mechanisms of bacterial meningitis in children.
